AB  - BACKGROUND
Fungi absorb and solubilize a broad spectrum of heavy metals such as vanadium (V), which makes them a main route of its entry into the biosphere. V as vanadate (V5+) is a potential medical agent due to its many metabolic actions such as interaction with phosphates in the cell, and especially its insulin-mimetic activity. Antidiabetic activity of V-enriched fungi has been studied in recent years, but the biological and chemical bases of vanadium action and status in fungi in general are poorly understood, with almost no information on edible fungi. 
METHODS
This manuscript gives a deeper insight into the interaction of V5+ with Coprinellus truncorum, an edible autochthonous species widely distributed in Europe and North America. Vanadium uptake and accumulation as V5+ was studied by 51V NMR, while the reducing abilities of the mycelium were determined by EPR. 31P NMR was used to determine its effects on the metabolism of phosphate compounds, with particular focus on phosphate sugars identified using HPLC.
RESULTS
Vanadate enters the mycelium in monomeric form and shows no immediate detrimental effects on intracellular pH or polyphosphate (PPc) levels, even when applied at physiologically high concentrations (20 mM Na3VO4). Once absorbed, it is partially reduced to less toxic vanadyl (V4+) with notable unreduced portion, which leads to a large increase in phosphorylated sugar levels, especially glucose-1-phosphate (G1P) and fructose-6-phosphate (F6P).
CONCLUSIONS
Preservation of pH and especially PPc reflects maintenance of the energy status of the mycelium, i.e., its tolerance to high V5+ concentrations. Rise in G1P and F6P levels implies that the main targets of V5+ are most likely phosphoglucomutase and phosphoglucokinase(s), enzymes involved in early stages of G6P transformation in glycolysis and glycogen metabolism. This study recommends C. truncorum for further investigation as a potential antidiabetic agent.

AB  - In recent years, versatile peroxidase (VP) has emerged as a promising enzyme for biotechnological applications, as it can oxidize lignin without the external mediators. To gain insights into the breakdown process of artificial lignin by VP, reaction between the two was studied. Degradation products were fractionated using ultrafiltration and analyzed by RP- high performance liquid chromatography with mass detection (HPLC-MS) chromatography. Four fractions were obtained based on their molecular sizes: >10, 3–10, 1–3, and <1 kDa. Interestingly, while VP did not significantly alter the yields of these fractions, the chromatograms revealed the presence of oligomers with different molecular weights (MWs) resulting from the enzymatic activity. The VP exhibits a dual role in its enzymatic activity: both degrading and synthesizing these oligomers. This was confirmed by principal component analysis (PCA). The positive correlations were found between certain oligomers (D1 and D2, D5 and D6, as well as between D7, D10, T2, and T4), suggesting their simultaneous degradation. On the other hand, a negative correlation was found between the monomer and some oligomers (D7, D10, T2, and T4), indicating the decomposition of these oligomers into monomers. These findings shed light on the intricate interplay between VP and artificial lignin, offering valuable insights for potential applications in lignin valorization.

AB  - (1) The nutritional quality of strawberry (Fragaria × ananassa Duch) fruits, among others, is largely maintained by the presence of soluble sugars and organic acids. As the primary products of photosynthesis, they are energy depots in plants, necessary for the construction of cell constituents, but also serve as precursors of aromatic compounds and signaling molecules. (2) In this study, fruits of 25 strawberry cultivars were qualitatively and quantitatively characterized concerning individual sugars and organic acids by HPLC, FT-ICR-MS, and MS imaging analysis. In addition, the total quality index (TQI), as a novel mathematical model, was used to compare all individual parameters evaluated to obtain a quantitative single score, as an indicator of overall fruit quality. (3) Regardless of a large number of cultivars and monitored parameters that were studded, several cultivars stood out in terms of selected primary metabolites, such as ‘Rumba’, ‘Jeny’, and ‘Sandra’, while the latter had the best TQI score. (4) Intercultivar variations in sugars and organic acids profiles, along with other bioactive compounds, should be considered for selection of promising cultivars with improved naturally occurring nutraceutical traits. Besides the search for a pleasant taste, increased awareness of healthy nutrition resulted in heightening consumer demand for high-quality fruit.

Quantitative traits of white cabbage cultivars in association with two biofertilisers - Abstract

AB  - Cabbage is one of the most important vegetables from Brassicaceae family to which 
biofertilisers is increasingly applied following the current trend in the cabbage organic 
farming. The main goal of our study was to examine whether solely applicated biofertilisers 
enhance head weight and other morphological parameters in cabbage. Experiment with three 
Bejo Zaden cultivars ('Farao' F1, 'Tiara' F1, and 'Excalibur' F1) were conducted under the open 
field conditions from April to June 2019. Two different microbiological fertilisers, Organic 
balance and FitoHelp, were applied foliar, six times during the vegetation period, with a 
battery sprayer. After harvest, several parameters were estimated: head weight, width and 
height, number of leaves, internal core height and width. The results showed the highest 
value of head weight (1264.89 g), head width (15.06 cm), head height (15.57 cm), number of 
leaves (22.56), internal core height (8.01 cm) and internal core width (3.51 cm) in cultivar 
'Excalibur' F1 compared to other two cultivars for fresh market. Application of both fertilisers 
led to increased levels of all tested parameters compared to control. Furthermore, fertiliser 
Organic balance showed the highest impact on all tested parameters, resulting in increasing 
value of head weight by 78.6%. This study revealed that mixed culture fertiliser showed 
greater impact in terms of head weight, number of leaves, and internal core width compared 
to fertiliser containing a single strain of bacteria. Accordingly, with an exception in head and 
internal core height, all examined factors showed statistically significant impact on tested 
parameters, including their interaction.

AB  - Cabbage is one of the most important vegetables from the Brassicaceae family to
which biofertilisers are increasingly applied following the current trend in cabbage
organic farming. The main goal of our study was to examine whether solely
applicated biofertilisers enhance head weight and other morphological parameters
in cabbage. Experiment with three Bejo Zaden cultivars ('Farao' F 1 , 'Tiara' F 1 , and
'Excalibur' F 1 ) were conducted under the open field conditions from April to June
2019. Two different microbiological fertilisers, Organic balance and FitoHelp,
were applied foliar, six times during the vegetation period, with a battery sprayer.
After harvest, several parameters were estimated: head weight, width and height,
number of leaves, internal core height and width. The results showed the highest
value of head weight (1264.89 g), head width (15.06 cm), head height (15.57 cm),
number of leaves (22.56), internal core height (8.01 cm) and internal core width
(3.51 cm) in cultivar 'Excalibur' F1 compared to other two cultivars for fresh
market. Application of both fertilisers led to increased levels of all tested
parameters compared to the control. Furthermore, fertiliser Organic balance
showed the highest impact on all tested parameters, resulting in an increasing value
of head weight by 78.6%. This study revealed that mixed culture fertiliser showed
a greater impact in terms of head weight, number of leaves, and internal core width
compared to fertiliser containing a single strain of bacteria. Accordingly, with
an exception in head and internal core height, all examined factors showed a
statistically significant impact on tested parameters, including their interaction.

AB  - Insulin resistance is a state where a normal amount of insulin can’t provoke an appropriate metabolic response. Insulin promotes membrane trafficking of the glucose transporter GLUT4 from the storage vesicles to the plasma membrane in white adipose tissue. Adipocytes use glucose for lipogenesis and store the energy as lipid droplets. If adipocytes are unable to uptake glucose, a chronic state of hyperglycemia is developed, with severe health consequences. Polyphenols are natural anti-inflammatory and antioxidant agents. Food rich in polyphenols has been suggested to exert an ameliorative effect on restoring insulin sensitivity, with the main identified target being AMPK1,2, one of the key sensors of intracellular energy. Here, we tested the effect of methanol extracts from three newly introduced strawberry (Fragaria x ananassa, Duch.) cultivars – 'Aprika', 'Sandra' and 'Quicky' on glucose metabolism in 3T3-F442A adipocytes. It was determined that 'Aprika' has the highest total phenolic content, relative to the other two cultivars. After 72-h exposure, none of the strawberry cultivars affected adipocyte cell growth significantly. Protein expression analysis of the differentiated adipocytes suggested 'Aprika', but not the other two cultivars, significantly increased the AMPK expression, as well as GLUT4, thus increasing glucose uptake. Strawberry extracts did not significantly affect the differentiation of adipocytes (SIRT1 and PPARγ), nor the fatty acid synthesis (ACC). Conclusively, the 'Aprika' methanol extract with high phenolic content exerts an ameliorative effect on glucose uptake, presumably through activation of the AMPK-dependent mechanism of GLUT4 trafficking. The systemic effects of the ‘Aprika’ cultivar should be further investigated. Implications of the research are decreased hyperglycemia in obese and diabetic patients, by the introduction of the 'Aprika' strawberry cultivar into everyday diet. References 1. AB  - The aim of this study was to compare newly released June-bearing
strawberry cultivars (‘Quicky’, ‘Sandra’, ‘Lofty’, ‘Nadja’ and ‘Aprica’) in terms of
their phenology, vegetative growth, productivity and fruit quality to identify their
potential for wider cultivation. A field study was conducted in a strawberry
plantation established in July 2020 in double rows on beds covered with black
polyethylene foil (Šid, Serbia). The cultivars were evaluated in 2021–2022 for their
flowering and ripening time, productivity, plant growth, biometrical and nutritional
fruit traits (soluble solids content – SSC, total acids – TAs, vitamin C, total
anthocyanins – TACY, total phenolics – TPC and total antioxidant capacity – TAC).
The cultivar ‘Quicky’ started to ripen earliest, while ‘Aprica’ was the latest in both
experimental years. The number of branch crowns per plant was significantly higher
in ‘Sandra’ and ‘Lofty’, whereby ‘Sandra’ had also the highest number of leaves per
rosette (41.5) in comparison with the other tested cultivars. The cultivar ‘Aprica’ was
superior in terms of productivity (1061 g/plant and 4.67 kg/m2
), fruit weight (29.9 g)
and fruit shape index (1.15). Contrary to this, ‘Nadja’ was the least productive
cultivar (608 g/plant and 2.68 kg/m2). The cultivars ‘Lofty’ and ‘Sandra’ showed the
highest SSC values and were also characterized by a considerably high level of TPC
(1.29 mg GAE eq g-1 FW) and TACY (24.4 mg pg-3-g eq 100 g-1 FW), respectively.
Variability among the tested cultivars could serve as an important criterion for the
selection of new high-performing cultivars for a given growing region.

AB  - In nature, woody substrates are usually degraded by entire communities of microorganisms, which are nowadays jeopardised by anthropogenic influence, making it important to define the functional specificity of every species. Two strains of autochthonous fungi from Serbian lowland forests (Pleurotus ostreatus Ser1 and Cyclocybe aegerita Ser1) have been investigated for their ability to degrade lignin substrates [oak sawdust, oak isolated cell walls, and synthetic dehydrogenative polymer (DHP)]. Measuring the activities of the enzymes involved in lignin degradation was coupled with detecting the HPLC profile of the phenolics in the fungal growth media, and the lignin loss. While Pleurotus ostreatus Ser1 appeared highly effective within a very short time span, Cyclocybe aegerita Ser1 failed to degrade lignin. This situation was supported by very high enzyme activities and the low presence of phenolics in the media of Pleurotus ostreatus Ser1, compared to very low enzyme activity and the high presence of phenolics in the media with Cyclocybe aegerita Ser1.

AB  - Maize is important to global food security, being one of the predominant cereals in human and domesticated livestock diets worldwide. Due to the increasing human population, it will be important to not only design cropping systems to increase maize yield and sustainability but also to improve the nutritional quality of maize edible tissues. To determine cropping system impacts on maize grain nutritional content, we sampled grain from conventional and organic maize varieties grown for three growing seasons using five cropping systems. We analyzed the grain using metabolic fingerprinting of methanol extracts with ultra-high performance liquid chromatography (UHPLC) coupled with mass spectrometry (MS), adopting both non-targeted and targeted approaches. The cropping systems are part of a long-term study at the Beltsville Agricultural Research Center in Beltsville, Maryland, and were a three-year conventional no-till rotation (NT), a three-year conventional chisel-till rotation (CT), a two-year organic rotation (Org2), a three-year organic rotation (Org3), and a six-year organic rotation (Org6). Each cropping system had been in place for at least 10 years, allowing specific cropping-system-induced alterations of soil edaphic and microbial properties. Non-targeted metabolic fingerprinting detected a total of 90 compounds, the majority of which were phenolics. Metabolic profiling was further targeted toward 15 phenolics, 1 phytohormone, 7 carbohydrates and 7 organic acids, which were quantified in the maize grain originating from the five cropping systems. Statistical analysis of this subset of quantitative data determined that cropping system can significantly influence levels of certain maize grain metabolites. However, natural impacts (growing year) were substantially greater than cropping system impacts, likely masking or over-riding some cropping system impacts. Additionally, maize cultivar genetics had greater impact than cropping system on the maize grain metabolome and was the greatest “managed” impact on the metabolite profiles. Results indicate that until natural environmental impacts on maize grain metabolite levels are understood and managed, the best approach to reliably increase maize grain nutritional quality is through development of maize cultivars with enhanced nutritional content that are robust to natural environmental influence.

AB  - The main goal of our study was to find an optimal combination of tested factors to achieve lettuce rich in bioactive compounds sustaining its pleasant taste. We examined three red and three green cultivars in a greenhouse using two microbiological fertilisers (EM Aktiv and Vital Tricho), and their combination. Plants were grown in three consecutive growing seasons (autumn, winter, and spring). Lactones accumulated in autumn, whereas phenolics’ concentration rose during winter. Red cultivars showed higher phenolics and lactone content, where chicoric acid and luteolin-7-glucoside were the most abundant in the ‘Gaugin’ winter trial. Lactucopicrin was the predominant lactone among tested cultivars with the highest value in the red cultivar ‘Carmesi’. Solely applicated, the fertiliser EM Aktiv and Vital Tricho led to significantly higher phenolic acid and dihydrolactucopicrin content, while combined, there were notably increased levels of all detected lactones. Application of single fertilisers had no effect on flavonoid content, while the combination even reduced it. A sensory analysis showed a negative correlation between overall taste and total sesquiterpene lactones, lactucopicrin, caffeoylmalic, and chlorogenic acid, indicating a less bitter taste with decreasing content of these compounds. Our findings indicate that the cultivar, fertiliser, and growing season jointly affected all of the tested parameters, highlighting the differences in the application of EM Aktiv, Vital Tricho, and their combination.

The major sesquiterpene lactone was lactucopicrin with the highest level found in cultivar ‘Carmesi’ (0.65 mg/g dry weight) in autumn with the combination of fertilisers. The concentrations of lactucin, dihydrolactucin and dihydrolactucopicrin ranged from 0.001-0.085, 0.003-0.015, and 0.001-0.056 mg/g dry weight, respectively, and were not found in all lettuce samples. Generally, red cultivars showed higher content of lactones compared to green. Application of Vital Tricho and the combination of fertilisers led to an increased level of lactucopicrin, in some cultivars, in the range of 77-800% compared to unfertilised plants. Higher levels of lactucopicrin were found in the autumn trial compared to spring and winter. The present study suggests that genotype, fertilisers and season jointly affected the quantity of sesquiterpene lactones with emphasis on Vital Tricho, and/or the combination of fertilisers.

AB  - Silicon (Si) is considered a beneficial element for plants, mostly accumulating in cell walls, where its location and content are primed by the chemistry and structure of lignin. It is unrevealed how Si interacts with the process of lignin formation in the CWs. We studied, in an in vitro system, the interaction of SiO2 with the peroxidase-catalyzed polymerization of a lignin monomer into the lignin model compound, imitating conditions of the last step of lignin formation. FTIR and fluorescence spectroscopy and microscopy showed that Si is bound to the final polymer, and the structure of the Si-DHP differs from pure DHP. Fluorescence spectroscopy showed that Si does not bind to the monomers, so Si probably inhibits the formation of the larger lignin fragments, as evidenced by HPLC-DAD, by binding to dimmers formed during DHP synthesis. The structural changes of the polymer are related to the changed proportion of the fractions of various MW. The enzyme catalyzing DHP synthesis was not inhibited by Si. HRP activity was increased in presence of Si except for 6 mM Si. This may indicate that the complex formed with Si and short oligomers activates the enzyme, and prevents the formation of the large fragments.

Нутритивне и биоактивне компоненте у плодовима једнородних и сталнорађајућих сорти јагодe

AB  - Поред шећера и витамина, плод јагоде (Fragaria×ananassa Duch) је важан извор и биоактивних једињења, као што су укупни феноли и антоцијани. Ова једињења истовремено дефинишу нутритивни и сензорички квалитет плода. У овим истраживањима испитиване су новије сорте јагоде , које су још увек у фази тестирања, и то три једнородне сорте ('Aprika', 'Sibilla', 'Vivaldi') и три сталнорађајуће сорте ('Albion', 'Capri', 'Irma') у којима су одређене концентрације индивидуалних шећера, органских киселина, витамина Ц, укупних фенола (TPC) и укупних антоцијана (TACY). Од једнородних сорти 'Vivaldi' се издвојила по садржају TACY (24.92 mg Pg-3-g eq/100 g свеже масе (FW)) и витамина Ц (46.7 mg/100 g FW), док је 'Sibilla' била доминантна у садржају глукозе и фруктозе (2.58 и 2.91 g/100 g FW, по редоследу), као и свих органских киселина детектованих помоћу HPLC. Од свих испитиваних параметара, једино је садржај TPC (1.41 mg GAE (gallic acid equivalents)/FW) био повећан код сорте 'Aprika' у односу на друге сорте. Од сталнорађајућих сорти 'Albion' се значајно издваја по високом садржају TACY (12.37 mg Pg-3-g eq/100 g FW) и витамина Ц (48.1 mg/100 g FW), док је 'Capri' била доминантнија у садржају фруктозе и глукозе (5.60 и 6.18 g/100 g FW, по редоследу), већине органских киселина, као и у садржају TPC (0.74 mg GAE/FW).  Са аспекта здравствене корисности плода, као перспективне сорте могу се препоручити  'Aprika' од једнородних и 'Capri' од сталнорађајућих сорти.

abstract = "Поред шећера и витамина, плод јагоде (Fragaria×ananassa Duch) је важан извор и биоактивних једињења, као што су укупни феноли и антоцијани. Ова једињења истовремено дефинишу нутритивни и сензорички квалитет плода. У овим истраживањима испитиване су новије сорте јагоде , које су још увек у фази тестирања, и то три једнородне сорте ('Aprika', 'Sibilla', 'Vivaldi') и три сталнорађајуће сорте ('Albion', 'Capri', 'Irma') у којима су одређене концентрације индивидуалних шећера, органских киселина, витамина Ц, укупних фенола (TPC) и укупних антоцијана (TACY). Од једнородних сорти 'Vivaldi' се издвојила по садржају TACY (24.92 mg Pg-3-g eq/100 g свеже масе (FW)) и витамина Ц (46.7 mg/100 g FW), док је 'Sibilla' била доминантна у садржају глукозе и фруктозе (2.58 и 2.91 g/100 g FW, по редоследу), као и свих органских киселина детектованих помоћу HPLC. Од свих испитиваних параметара, једино је садржај TPC (1.41 mg GAE (gallic acid equivalents)/FW) био повећан код сорте 'Aprika' у односу на друге сорте. Од сталнорађајућих сорти 'Albion' се значајно издваја по високом садржају TACY (12.37 mg Pg-3-g eq/100 g FW) и витамина Ц (48.1 mg/100 g FW), док је 'Capri' била доминантнија у садржају фруктозе и глукозе (5.60 и 6.18 g/100 g FW, по редоследу), већине органских киселина, као и у садржају TPC (0.74 mg GAE/FW).  Са аспекта здравствене корисности плода, као перспективне сорте могу се препоручити  'Aprika' од једнородних и 'Capri' од сталнорађајућих сорти.",

AB  - The most popular cultivated strawberry is the dessert strawberry (Fragaria×ananassa Duch.)
from which breeders have created an abundance of new improved cultivars with specific agronomic, qualitative and sensorial characteristics. In this study, two newly introduced strawberry
cultivars ‘Joly’ and ‘Sibilla’ were compared with their mutual parent and commercial cultivar ‘Clery’
in terms of individual phenolics, sugars, and organic acids content. All forms of free pelargonidin
were significantly higher in ‘Joly’ and ‘Sibilla’ (17.6 and 11.3 µg/g FW, respectively) compared to
‘Clery’ (3.4 µg/g FW), in which cyanidin 3-glucoside is the dominant (6.2 µg/g FW). Quercetin
3-O-glucuronide values were higher in ‘Sibilla’ and ‘Joly’ (17.4 and 11.6 µg/g FW, respectively) than
in ‘Clery’ (2.6 µg/g FW). The highest values of p-coumaric acid were detected in ‘Sibilla’ (6.2 µg/g
FW). Fructose content (3.1 g/100g FW) was the highest in ‘Joly’, sucrose content (0.8 g/100g FW)
in ‘Sibilla’, while glucose content was similar in all three cultivars. The highest value of fumaric and
citric acid (13.1 µg/g and 6.2 mg/g FW, respectively) was detected in ‘Sibilla’, while malic and shikimic acid had similar values in all three cultivars. Two newer strawberry cultivars ‘Joly’ and ‘Sibilla’ generally had higher levels of analyzed bioactive compounds compared to their common parent ‘Clery’. Given that sensorial and nutritional quality of strawberry fruit is significantly affected by the ratio of sugars and organic acids, as well as their combination with phenolic compounds, the established better fruit quality of newer cultivars indicated their significant potential for commercial
cultivation.

Rich in Phenolics—Strong Antioxidant Fruit? Comparative Study of 25 Strawberry Cultivars

AB  - Phenolic compounds of 25 newly introduced strawberry cultivars were profiled using spectrophotometry, electron paramagnetic resonance (EPR) spectroscopy, and high-performance liquid chromatography-mass spectrometry. Total phenolic and anthocyanin content (TPC and TACY, respectively), as well as vitamin C, and concentrations of individual phenolic compounds in fruits were evaluated to identify the most promising cultivars according to their phenolic profile. The highest values of TPC, TACY, and vitamin C were recorded in ´Premy´ (1.53 mg eq GA g-1 FW), ´Sandra´ (30.60 mg eq Pg-3-g 100 g-1 FW) and ´Laetitia´ (56.32 mg 100 g-1 FW), respectively. The DPPH and •OH radicals scavenging activity of fruit methanolic extracts was estimated using EPR spectroscopy. All cultivars are almost uniformly effective in the scavenging of •OH radical, while ´Tea´, ´Premy´, and ´Joly´ were marked as highly potent cultivars (over 70%) in terms of DPPH-antiradical activity. Specific peroxidase activities were the highest in ´Garda´, ´Federica´, and ´Rumba´ (0.11, 0.08, and 0.06 U mg-1 prot, respectively). ´Laetitia´, ´Joly´, ´Arianna´, ´Tea´, and ´Mila´ cultivars were distinguished from others as the richest concerning almost all flavonoids and phenolic acids, including some other parameters of bioactivity. These cultivars could be recommended to consumers in terms of fruit as a functional food.

Productivity and fruit quality of 'Clery' strawberry affected by planting density in a soilless growing system

AB  - This study illustrates the differences in yield components, morphometric and nutritional fruit properties of the strawberry cultivar 'Clery' affected by planting density in a soilless growing system. Cold stored frigo plants were planted at the beginning of August 2018 in bags filled with 20 L substrate mixture which were placed within styrofoam containers on a 1.2 m high tabletop. The effect of two planting densities, 10 and 14 plants m(-2), on mentioned parameters was monitored during 2019 in a greenhouse from the 'Euro Fruit' company in Kupinovo, Serbia. Results showed that the harvest window was similar in both applied planting densities and it lasted around one month (from April 20 to May 20, 2019). Contrary to this, a significantly higher number of fruits plant(-1), yield plant(-1) and average fruit weight was registered in the density of 10 plants m(-2) (27.8; 545.4 g plant(-1); 20.0 g, respectively). Higher planting density of 14 plants m(-2) significantly increased concentrations of glucose, citric, malic and shikimic acids in the fruit (20.3 g kg(-1); 5.41 mg g(-1); 2.23 mg g(-1) and 20.13 mu g g(-1), respectively). Soluble solids, fructose and vitamin C content in fruit were not affected by the planting density. These results indicate that the use of a planting density of 10 plants m(-2) could be considered more suitable in a soilless production system of 'Clery' with optimal yields plant(-1) and m(-2) despite positive effects on some morphometric and chemical fruit traits at the density of 14 plants m(-2).

A comparison of major taste- and health-related compounds among newly released Italian strawberry cultivars

AB  - Strawberry (Fragaria x ananassa Duch.) is one of the most popular edible fruit worldwide of which consumption constantly increases with the development of new cultivars available at almost all season. Besides being delicious, strawberry fruits are also an important source of a wide variety of nutritive compounds such as sugars, vitamins and minerals, as well as bioactive compounds such as phenolics. Nutritive composition of sugar, organic acid, and phenolic compounds is very diversely distributed in different strawberry cultivars. Considering that sugars and organic acids significantly affect the sensorial quality of strawberry fruit, their optimal ratio combined with phenolic compounds should be accomplished in order to obtain the high quality fruit. Fruit of six strawberry cultivars newly introduced from Italy ('Aprika', 'Sibilla', 'Garda', 'Premy', ' Jeny', 'Laetitia') were characterized in relation to the total phenolic content and antioxidant capacity, vitamin C, soluble solids content, titratable acidity, as well as concentration of individual sugars and organic acids. 'Jeny' was the dominant strawberry cultivar in terms of glucose (4.34 g 100 g(-1) fresh weight - FW), fructose (4.68 g 100 g(-1) FW), malic (2.93 mg g(-1) FW) and shikimic (21.55 mu g g(-1) FW) acid content, while 'Laetitia' was distinguished among all other cultivars regarding total antioxidant capacity (3.99 mg AsA eq g(-1) FW), citric acid (6.86 mg g(-1) FW) and vitamin C (56.32 mg 100 g(-1) FW). Hence 'Laetitia', as the best ranking cultivars according to the content of nutritive compounds with proven bioactive effects, could be recommended for consumption in relation to the consumer preferences concerning human health and wellbeing.

Considering that sugars and organic acids significantly affect the sensorial quality of strawberry fruit, their optimal ratio combined with phenolic compounds should be accomplished in order to obtain the high quality fruit. Fruit of six strawberry cultivars newly introduced from Italy ('Aprika', 'Sibilla', 'Garda', 'Premy', ' Jeny', 'Laetitia') were characterized in relation to the total phenolic content and antioxidant capacity, vitamin C, soluble solids content, titratable acidity, as well as concentration of individual sugars and organic acids. 'Jeny' was the dominant strawberry cultivar in terms of glucose (4.34 g 100 g(-1) fresh weight - FW), fructose (4.68 g 100 g(-1) FW), malic (2.93 mg g(-1) FW) and shikimic (21.55 mu g g(-1) FW) acid content, while 'Laetitia' was distinguished among all other cultivars regarding total antioxidant capacity (3.99 mg AsA eq g(-1) FW), citric acid (6.86 mg g(-1) FW) and vitamin C (56.32 mg 100 g(-1) FW). Hence 'Laetitia', as the best ranking cultivars according to the content of nutritive compounds with proven bioactive effects, could be recommended for consumption in relation to the consumer preferences concerning human health and wellbeing.",

The aim of this study was to examine the effect of genotype, microbiological fertilisers and season on the rosette fresh weight and components of antioxidant activity. Six cultivars (green 'Kiribati', 'Aleppo', 'Aquino' and red 'Murai', 'Carmesi', 'Gaugin') were grown in a greenhouse experiment during three consecutive seasons (autumn, winter and spring) with application of microbiological fertilisers (EM Aktiv, Vital Tricho and their combination). Green cultivars showed higher fresh weight than red in spring and winter. Green cultivar 'Aleppo' showed the highest fresh weight in control during spring. Microbiological fertilisers led to increased fresh weight in autumn. Mainly, red cultivars showed higher quality parameters compared to green (total antioxidant capacity, total phenolic content, total carotenoids and POD activity). Red cultivar 'Carmesi' showed the highest total antioxidant capacity, total carotenoids, and total phenolic content in winter with combination of fertilisers, Vital Tricho, and in control. The present study suggested that genotype, fertilisers and season jointly influenced quantity and quality parameters with emphasis on Vital Tricho, and/or combination of fertilisers.

AB  - The UV-B represents the minor fraction of the solar spectrum, while UV-C is not contained in natural solar radiation, but both radiation types can cause damaging effects in plants. Cell walls (CWs) are one of the targets for external stressors. Juvenile P. omorika trees were treated either with 21 day-high doses UV-B or with 7 day-UV-C in open-top chambers. Using spectroscopic and biochemical techniques, it was shown that the response to UV radiation includes numerous modifications in needle CW structure: relative content of xylan, xyloglucan, lignin and cellulose decreased; cellulose crystallinity changed; yield of lignin monomers with stronger connection of C=C in side chain with the ring increased; re-distribution of inter- and intra-polymer H-bonds occurred. The recovery was mediated by an increase in the activities and changes in isoform profiles of CW bound covalent peroxidases (POD) and polyphenol oxidases (PO) (UV-B), and ionic POD and covalent PO (UV-C). A connection between activities of specific POD/PO isoforms and phenolic species (m- and p-coumaric acid, pinoresinol and cinnamic acid derivatives) was demonstrated, and supported by changes in the sRNA profile. In vivo fluorometry showed phenolics accumulation in needle epidermal CWs. These results imply transversal connections between polymers and changed mechanical properties of needle CW as a response to UV. The CW alterations enabled maintenance of physiological functions, as indicated by the preserved chlorophyll content and/or organization. The current study provides evidence that in conifers, needle CW response to both UV-B and UV-C includes biochemical modifications and structural remodeling.

Spatial distribution of apoplastic antioxidative constituents in maize root

AB  - Apoplastic antioxidative constituents (enzymes, primary and secondary metabolites, ROS) from different root zones of hydroponically grown maize (Zea mays L.) were investigated using a noninvasive isolation procedure: filter strip method. Filter strips were placed at specific positions on the root surface: apical zone (tip) and basal zone (base) to absorb apoplastic fluid. Three major classes of low-weight metabolites (organic acids, sugars, and phenolics) have been identified by HPLC-ECD. The longitudinal distribution of sugars and organic acids had the same pattern: higher concentration in the tip than the base, while it was vice versa for phenolics. The specific activities of guaiacol peroxidase, superoxide dismutase, and ascorbate peroxidase were higher in the apoplastic fluid from the root base than the tip, and their different isoforms were separated by isoelectric focusing. Electron paramagnetic resonance (EPR) spectroscopy coupled with the spin-trapping method using DEPMPO showed a persistent generation of hydroxyl radical in the root tip. In vivo EPR imaging of the whole maize root with membrane-permeable and impermeable aminoxyl spin-probes, enabling real-time detection of ROS formation within and outside the membranes, demonstrated ROS accumulation on the root surface, while endodermis and central cylinder were ROS free. For the first time in plant research, 2D EPR images enabled the direct demonstration of site-specific free radical production along the root. Highly sensitive analytical techniques combined with the filter strips, as a non-invasive tool, have increased our knowledge of metabolic processes occurring in the apoplast and their spatial-temporal changes in small regions of the intact root tissue.

However, the effect of French Marigold volatiles on potato metabolism, physiology and induced defense is unknown. Thus, a microarray transcriptome analysis was performed to study the effects of French Marigold essential oil (EO) on laboratory-grown potato. After 8 h of exposure to EO, with gas chromatography/mass spectrometry (GC/MS)-detected terpinolene and limonene as dominant compounds, 2796 transcripts were differentially expressed with fold change >2 compared to expression in controls. A slightly higher number of transcripts had suppressed expression (1493 down- vs. 1303 up-regulated). Since transcripts, annotated to different photosynthesis-related processes, were mostly down-regulated, we selected a set of 10 genes involved in the leaf starch metabolism pathway, and validated microarray patterns using quantitative reverse transcription polymerase chain reaction (RT-qPCR). Except for decreased synthesis and induced decomposition of starch granule in leaves, 8 h long EO exposure slightly elevated the accumulation of sucrose compared to glucose and fructose in subjected potato plants. An in vitro feeding bioassay with Colorado potato beetle showed that EO-induced alternations on transcriptional level and in the sugars' metabolism caused the enhancement of feeding behavior and overall development of the tested larvae. Results of comprehensive analysis of transcriptional responses in potato exposed to French Marigold EO provide a basis for further elucidation of molecular mechanisms underlying eco-physiological interactions in companion planting cropping systems.

AB  - Phosphinothricin (PPT) is one of the most widely used herbicides. PTT targets glutamine synthetase (GS) activity in plants, and its phytotoxicity is ascribed to ammonium accumulation and reactive oxygen species bursts, which drives rapid lipid peroxidation of cell membranes. In agricultural fields, PPT is extensively sprayed on plant foliage; however, a portion of the herbicide reaches the soil. According to the present study, PPT absorbed via roots can be phytotoxic to Arabidopsis, inducing more adverse effects in roots than in shoots. Alterations in plant physiology caused by 10 days exposure to herbicide via roots are reflected through growth suppression, reduced chlorophyll content, perturbations in the sugar and organic acid metabolism, modifications in the activities and abundances of GS, catalase, peroxidase, and superoxide dismutase. Antagonistic interaction of Nepeta rtanjensis essential oil (NrEO) and PPT, emphasizes the existence of complex control mechanisms at the transcriptional and posttranslational level, which result in the mitigation of PPT-induced ammonium toxicity and in providing more efficient antioxidant defense of plants. Simultaneous application of the two agents in the field cannot be recommended; however, NrEO might be considered as the PPT post-treatment for reducing harmful effects of herbicide residues in the soil on non-target plants.

AB  - We present changes in Tacitus bellus antioxidative system that specifically correspond to subsequent phases of hemibiotroph Fusarium verticillioides infection revealed by histological analysis. T. bellus response to spore germination 6 h post inoculation (hpi), manifested as first oxidative burst, was characterized by transient decrease in malondialdehyde (MDA) content, transient increase in catalase (CAT), low level of superoxide dismutase (SOD) and peroxidase (POD) activity, as well as with transient decrease in total antioxidant capacity (TAC), total phenol content (TPC) and phenylalanine ammonium lyase activity (PAL), and no changes in polyphenol oxidase (PPO) activity, or phenolic profile. During the biotrophic phase of F. verticillioides infection, characterized by hyphae spread intercellularly in epidermal and mesophyll tissue, the host antioxidative system was suppressed. The transition to necrotrophic phase of F. verticillioides infection (inter- and intracellular colonization and sporulation), occurred 3-4 days post inoculation (dpi). During the necrotrophic phase, 5-7 dpi, slowed progression of colonization of T. bellus mesophyll cells occurred and it coincided with sharp increase in MDA content and CAT, SOD and POD activities, but the drop in TAC, TPC content, and PPO activity, as well as the production of phytotoxin fusaric acid. Presented results add to the knowledge of events and mechanisms related to the transition from biotrophy to necrotrophy in F. verticillioides.

In plants, phenolamides are involved in biotic stress response. The data on variations of phenolamides in AFB1-containing seeds are limited. RESULTS Five polyamine conjugates, including two spermidine and three putrescine conjugates, were tentatively identified in methanolic extracts, using HPLC-DAD-MS. The ratio of putrescine to spermidine conjugates changed with increasing AFB1 concentration in a logistic dose-response manner, with a ratio of below 1 up to a concentration of 51.51 mu g kg(-1), and approximately 2.54 and 3 at higher concentrations of 177.4 and 308.13 mu g kg(-1), respectively. The observed variations of the total antioxidant activity and the total phenolic content may support this biphasic behaviour of the seeds against AFB1 stress. CONCLUSIONS The obtained data are a contribution to the understanding of the roles of polyamine conjugates in seed defence to increasing AFB1 concentrations. According to our knowledge, this study reports for the first time the biphasic response of maize seeds to increasing AFB1 contamination level, comprising the induction of polyamine conjugate accumulation and variation in the ratio of conjugates. This dose-response relationship may provide useful information in the field of agricultural and food chemistry as an indicator of AFB1 contamination level and, hence, for selecting an appropriate seed quality.

AB  - Phenolic compounds of 12 strawberry cultivars were profiled using spectrophotometry, spectrofluorometry, and high-performance liquid chromatography-mass spectrometry, coupled with multivariate regression analysis. Total phenolic content, total anthocyanin content, and total antioxidant capacity (TPC, TACY, and TAC, respectively) and concentrations of individual phenolics were evaluated, and the multivariate statistic was employed to identify the most promising cultivars based on their phenolic content. According to the principal component analysis, TAC was strongly correlated with the TPC (0.81), pointing out its importance in overall antioxidant activity. 'Joly', 'Laetitia', and 'Asia' cultivars were distinguished from others as the richest in concerning identified anthocyanins, almost all flavonoids and phenolic acids. Multivariate curve resolution with alternating least squares indicated the presence of two main types of fluorophores assigned to anthocyanins and phenolics, in which emission spectral ratios also showed the highest values in the referred cultivars. These cultivars could be recommended to consumers in terms of the health functionality of fruit.
