AB  - Two Balkan Peninsula endemics, Nepeta rtanjensis and N. argolica subsp. argolica, both characterized by specialized metabolite profiles predominated by iridoids and phenolics, are differentiated according to the stereochemistry of major iridoid aglycone nepetalactone (NL). For the first time, the present study provides a comparative analysis of antimicrobial and immunomodulating activities of the two Nepeta species and their major iridoids isolated from natural sources-cis,trans-NL, trans,cis-NL, and 1,5,9-epideoxyloganic acid (1,5,9-eDLA), as well as of phenolic acid rosmarinic acid (RA). Methanol extracts and pure iridoids displayed excellent antimicrobial activity against eight strains of bacteria and seven strains of fungi. They were especially potent against food-borne pathogens such as L. monocytogenes, E. coli, S. aureus, Penicillium sp., and Aspergillus sp. Targeted iridoids were efficient agents in preventing biofilm formation of resistant P. aeruginosa strain, and they displayed additive antimicrobial interaction. Iridoids are, to a great extent, responsible for the prominent antimicrobial activities of the two Nepeta species, although are probably minor contributors to the moderate immunomodulatory effects. The analyzed iridoids and RA, individually or in mixtures, have the potential to be used in the pharmaceutical industry as potent antimicrobials, and in the food industry to increase the shelf life and safety of food products.

Antimicrobial composite polymers as potential agents in chronic wound healing

AB  - Biofilm-forming bacteria are recognized as a major impediment to wound healing. The aim of this study was to improve design of the formulations used for care of biofilm-infected wounds. Since lignins and lignin-derived compounds are known to have antibacterial properties especially efficient against biofilm-forming pathogens, the gel and membrane type of materials of enzymatically synthesized lignin model compound, dehydrogenative polymer of coniferyl alcohol (DHP) with alginate (Alg) and bacterial cellulose (BC) synthesized by culture Komagataeibacter intermedius IMBG180, were composed. The hydrogels were tested on Gram-negative P. aeruginosa (ATCC 27853) and S. typhimurium (ATCC 13311), Gram-positive bacteria L. monocytogenes (NCTC 7973) and S. aureus (ATCC 6538), as well as S. aureus #1*, S. aureus #2*, P. aeruginosa* and Serratia sp.* clinical human isolates obtained from patients’ wounds. DHP-Alg showed activity against all ofthe tested bacteria with minimal inhibitory concentrations (MICs) of 0.002–0.45 mg/mL and minimal bactericidal concentrations (MBCs) of 0.004–0.90 mg/mL. L. monocytogenes was the most susceptible. DHP-BC exhibited antibacterial activity on all tested bacteria, with exception of P. aeruginosa and S. aureus #1*. MICs and MBCs were in range of 0.22-0.42 mg/mL and 0.22-0.88 mg/mL, respectively. DHP-Alg gel and DHP-BC membranes, after initial release in the first hour of 34% and 19.26% of DHP, respectively, allowed the slow and continuous release of DHP in PBS solution. After 24 hours the amount of released antimicrobial substance from DHP-Alg and DHP-BC were 42.13% and 46.2%, respectively. Based on all obtained results, both hydrogels have the potential to be used in medical treatments as antibacterial agents.

AB  - Lignins and lignin-derived compounds are known to have antibacterial properties. The wound healing agents in the form of dressings produce faster skin repair and decrease pain in patients. In order to create an efficient antimicrobial agent in the form of dressing in the treatment of chronic wounds, a composite hydrogel of bacterial cellulose (BC) and dehydrogenative polymer of coniferyl alcohol (DHP), BC-DHP, was designed. Novel composite showed inhibitory or bactericidal effects against selected pathogenic bacteria, including clinically isolated ones. The highest release rate of DHP was in the first hour, while after 24 h there was still slow release of small amounts of DHP from BC-DHP during 72 h monitoring. High-performance liquid chromatography coupled with mass-spectrometry showed that BC-DHP releases DHP oligomers, which are proposed to be antimicrobially active DHP fractions. Scanning electron microscopy and atomic force microscopy micrographs proved a dose-dependent interaction of DHP with BC, which resulted in a decrease of the pore number and size in the cellulose membrane. The Fourier-transform infrared absorption spectra of the BC-DHP showed that DHP was partly bound to the BC matrix. The swelling and crystallinity degree were dose-dependent. All obtained results confirmed BC-DHP composite as a promising hydrogel for wounds healing.

AB  - Anti-biofilm activity of three anthocyanidins (pelargonidin, cyanidin and delphinidin) was evaluated for the first time at in vitro conditions. All the compounds reduced the formation of Pseudomonas aeruginosa PAO1 biofilm at low sub-MIC (0.125 MIC) with delphinidin (c 56.25g/mL) being the most active (43%). In comparison, ampicillin (c 93.75g/mL) and streptomycin (c 21.25g/mL) (used as positive controls) were considerably less effective at the same sub-MIC (8 and 12%, respectively). Furthermore, at 0.5 MIC (c 225g/mL) this anthocyanidin molecule partly reduced the bacterial protrusions. However, no any of the aforementioned compounds inhibited the production of pyocyanin by the bacterial strain P. aeruginosa PAO1. Taken all together, the delphinidin scaffold could be taken into consideration for the design of the novel and more effective anti-biofilm agents inspired by the anthocyanidins.

AB  - Nowadays bacterial resistance to known antibiotics is a serious health problem. In order to achieve more efficient treatment, lately there is an effort to find new substances, such as certain biomaterials, that are non-toxic to humans with antibiotic potential. Lignins and lignin-derived compounds have been proposed to be good candidates for use in medicine and health maintenance. In this study, the antibacterial activity of the lignin model polymer dehydrogenate polymer (DHP) in alginate hydrogel (Alg) was studied. The obtained results show that DHP-Alg has strong antimicrobial activity against several bacterial strains and biofilms and does not have a toxic effect on human epithelial cells. These results strongly suggest its application as a wound healing agent or as an adjunct substance for wound treatments.

AB  - The antimicrobial activity of five crude extracts of the freshwater bryozoan Pectinatella magnifica (Leidy, 1851) was evaluated in vitro for the first time. P. magnifica acetone extract exhibited the highest antibacterial activity (minimum inhibitory concentrations (MIC) 0.004-0.350mg/mL and MBC 0.007-0.500mg/mL), while its methanol extract showed the most promising antifungal activity (MIC 0.03-0.12mg/mL and MFC 0.06-0.25mg/mL). Furthermore, at a concentration of 0.25 MIC, the methanol extract reduced biofilm formation of the bacterial strain Pseudomonas aeruginosa PAO1 in a considerable extent (59.14%). FTIR spectra of the most active extracts indicate the presence of carbonyl compounds, long-chain alcohols and/or sterols. According to the experimental data obtained, P. magnifica methanol extract may be considered as a good resource of novel natural products with potent antibiofilm activity against the bacterium well known for its resistance. AB  - The antibiofilm and possible antiquorum sensing effects against the strain Pseudomonas aeruginosa PAO1 of five crude extracts of the freshwater bryozoan Hyalinella punctata (Hancock, 1850) were evaluated in vitro for the first time. H. punctata ethyl acetate extract (HpEtAc) exhibited the highest antibiofilm activity reducing the biofilm formation of P. aeruginosa PAO1 in the range of 80.63-88.13%. While all tested extracts reduced the twitching motility of the aforementioned bacterial strain, HpEtAc showed to be the most effective. Finally, at a concentration of 0.5 MIC, the same extract mostly inhibited the production of pyocyanin by P. aeruginosa PAO1 (71.53%). In comparison both with the positive controls used (streptomycin and ampicillin, 67.13 and 69.77%, respectively), HpEtAc was found to inhibit pyocyanin in a higher extent. An extensive chemical characterisation of this particular extract may result in isolation and identification of novel lead compounds targeting P. aeruginosa, an opportunistic human pathogen. AB  - This work extends in vitro screening of antimicrobial activity of avarol, the marine natural product firstly isolated from the Mediterranean sponge Dysidea avara. Its anticandidial activity was evaluated by microdilution method against eight Candida strains, two ATCC and six clinical ones. At a different extent this compound was proven to be active against all the strains tested (MIC 0.8-6.0g/mL and MFC 1.6-12.0g/mL, respectively). According to the best of our knowledge, this is the first report on avarol activity towards any yeast strain which may be of relevance for Alzheimer's disease. Indeed, avarol derivatives showing moderate AChE activity should be screened for anticandidial activity both in vitro and in vivo. [GRAPHICS] .

In vitro anti-quorum sensing activity of phytol

AB  - Anti-quorum sensing activity of the diterpene phytol was evaluated in vitro for the first time. This compound (at three sub-MIC concentrations - 0.5, 0.25 and 0.125 MIC, respectively) reduced the formation of Pseudomonas aeruginosa PAO1 biofilm in the range of 74.00-84.33% exhibiting higher activity than the both positive controls used, streptomycin and ampicillin. Phytol (0.5 MIC) also effectively reduced P. aeruginosa twitching and flagella motility. Indeed, the bacteria treated were incapable of producing a twitching zone and had almost round, smooth and regular colony edges. Finally, the tested compound (0.5 MIC) exhibited good P. aeruginosa pyocyanin inhibitory activity (51.94%) practically to the same extent as streptomycin (52.09%). According to the experimental data obtained, this phytol property may inspire design of medical foods targeting P. aeruginosa quorum sensing activity.

This compound (at three sub-MIC concentrations - 0.5, 0.25 and 0.125 MIC, respectively) reduced the formation of Pseudomonas aeruginosa PAO1 biofilm in the range of 74.00-84.33% exhibiting higher activity than the both positive controls used, streptomycin and ampicillin. Phytol (0.5 MIC) also effectively reduced P. aeruginosa twitching and flagella motility. Indeed, the bacteria treated were incapable of producing a twitching zone and had almost round, smooth and regular colony edges. Finally, the tested compound (0.5 MIC) exhibited good P. aeruginosa pyocyanin inhibitory activity (51.94%) practically to the same extent as streptomycin (52.09%). According to the experimental data obtained, this phytol property may inspire design of medical foods targeting P. aeruginosa quorum sensing activity.",

AB  - The antiradical activity of phytol was evaluated by electron paramagnetic resonance towards hydroxyl radical (root OH), superoxide anion radical (), methoxy radical (root CH2OH), carbon-dioxide anion radical (), as well as towards nitric-oxide radical (root NO) and 2,2-diphenyl-1-picrylhydrazyl (root DPPH) radical. It reduced the production of all tested radicals showing more promising activity against , root CH2OH and root DPPH radicals (56%, 50% and 48%, respectively) in comparison with root NO, and root OH radicals (38%, 23% and 15%, respectively). The antimicrobial activity of phytol was evaluated by the microdilution method against eight bacterial and eight fungal strains. To varying degrees, it was proven to be active against all tested bacteria and fungi (MIC 0.003-0.038mg/mL and MBC 0.013-0.052mg/mL, MIC 0.008-0.016mg/mL and MFC 0.090-0.520mg/mL, respectively). According to the obtained results, medical foods containing phytol may support development of new therapies for heart disease.

AB  - The anti-quorum sensing activities towards the bacterium Pseudomonas aeruginosa PA01 (pyocyanin production, biofilm formation and twitching and flagella motility) of two crude extracts (methanol and acetone) of the freshwater sponge Ochridaspongia rotunda (Arndt, 1937) were evaluated in vitro for the first time. Both extracts demonstrated P. aeruginosa pyocyanin inhibitory activity, reducing its production for 49.90% and 42.44%, respectively. In addition, they both showed higher anti-biofilm activity (48.29% and 53.99%, respectively) than ampicillin (30.84%). Finally, O. rotunda extracts effectively reduced twitching and flagella motility of P. aeruginosa. Taken all together, these results suggest that endemic sponge species from the oldest lake in Europe may offer novel bioactive natural products with promising medicinal potential towards P. aeruginosa infections.

AB  - The antimicrobial activity of five crude extracts (aqueous, methanol, ethyl acetate, acetone and methylene chloride) of the freshwater sponge Ochridaspongia rotunda (Arndt, 1937) was evaluated in vitro by using microdilution method against eight bacterial and eight fungal strains for the first time. The extracts were proven to be active in varying degrees against all the bacteria and fungi tested. O. rotunda methanol extract exhibited the highest antibacterial activity (minimum inhibitory concentration (MIC) 7.5-15.0 mu g/mL and minimum bactericidal concentration 15-30 mu g/mL), while its acetone extract exhibited the most promising antifungal activity (MIC 7.5-45.0 mu g/mL and minimum fungicidal concentration 15-60 mu g/mL). The extracts were more effective against the bacteria and fungi screened compared with the positive controls (streptomycin and ampicillin for bacteria and bifonazole and ketoconazole for fungi, respectively). According to the experimental data obtained, this deepwater sponge species may be considered as a gold mine of new antimicrobial substances with significant and broad-range activity.

AB  - This work extends in vitro screening of antimicrobial activity of the sesquiterpene hydroquinone avarol, a main secondary metabolite of the Mediterranean sponge species Dysidea avara. The antimicrobial activity was in part evaluated by microdilution method against selected bacterial and fungal strains. Additionally, the screening included evaluation of anti-quorum sensing (anti-QS) effects. At a different extent avarol was proven to be active against all the microorganisms tested (MIC 0.002-0.008 mg/mL and MBC 0.004-0.016 mg/mL; MIC 0.004-0.015 mg/mL and MFC 0.008-0.030 mg/mL; respectively). This marine natural product also showed moderate anti-QS effects, reducing Pseudomonas aeruginosa PAO1 biofilm formation (75%), its twitching and protrusions motilities as well as pyocianin production (39%). According to the best of our knowledge, this is the first report both on avarol anti Gram-negative bacterial activity and anti-QS effects.

The antiradical potential of the essential oil was evaluated using hydroxyl radical (center dot OH) generated in Fenton reaction. The meat preserving potential of essential oil from Satureja horvatii was investigated against L monocytogenes. Essential oil successfully inhibited development of L monocytogenes in pork meat. Sensorial evaluation on flavor and color of meat was performed. The color and flavor of meat treated with essential oil improved after 4 days of storage. S. horvatii essential oil can act as a potent inhibitor of food spoiling microorganisms, in meat products and also can be a useful source of natural antioxidants.

AB  - The antimicrobial activity of the moss Rhodobryum ontariense tea was evaluated by microdilution method against eight bacterial (Bacillus cereus, Enterobacter cloacae, Escherichia coli, Listeria monocytogens, Micrococcus flavus, Pseudomonas aeruginosa, Salmonella typhimurium and Staphylococcus aureus) and eight fungal species (Aspergillus fumigatus, Aspergillus niger, Aspergillus ochraceus, Aspergillus versicolor, Candida albicans, Penicillium funiculosum, Penicillium ochrochloron and Trichoderma viride). The investigated water extract was proven to be active against all the bacteria and majority of the fungi tested, but to varying degrees. E. cloacae was the most sensitive bacterium (MIC 0.62 mg/mL and MBC 2.50 mg/mL while C. albicans was the most sensitive micro fungus (MIC 0.20 mg/mL and MFC 1.00 mg/mL). On the other hand, at tested concentration the extract did not show any activity against the fungi A. fumigatus and A. niger. These findings indicated a good potential of R. ontariense tea in searching for new antifungals against C. albicans, the fungus which have become a cause of major health concerns in hospital due to hospital-acquired infections.

AB  - The aim of this study was to examine chemical composition and biological activity of Gaultheria procumbens L essential oil (EO) against food spoilage and oral microorganisms. The components of EO were identified by GC-MS. Antimicrobial activity was determined against food spoilage (five bacteria and six fungal species) and oral microorganisms (eight bacteria and thirty two fungal species) by microdilution and microplate biofilm assay, antioxidant activity was tested using the persistent free radical 2,2-diphenyl-1-picryl hydrazyl (DPPH), while antiradical activity was examined by fluorescence spectroscopy and electron paramagnetic resonance spectroscopy (EPR). GC-MS analysis showed that methyl salicylate (96.90%) was the main component of the oil. Essential oil inhibited the growth of all microorganisms tested, i.e. food and oral bacteria and fungi, respectively (MIC 0.18-3.00 mg/ml and MBC 1.25-4.00 mg/ml; MIC 0.73-5.00 mg/ml and MFC 2.92-26.67 mg/ml); The oil effectively inhibited the biofilm formation of oral Streptococcus mutans and Candida albicans as well (MIC 25.00 MBC 50.00 mg/ml; MIC 12.50, MFC 50.00 mg/ml). In addition, oil exhibited a dose-dependent DPPH-radical-scavenging activity with IC50 value of 30.61 mg/ml. The specific fluorescence probe 2-[6-(4'-amino)phenoxy-3H-xanten-3-on-9-yl] benzoic acid (APP) and the the spin trap 5-(Diethoxyphosphoryl)-5-methyl-1-pyrroline-N-oxide (DEPMPO), capable for simultaneous detection of different free radical specie were used in antiradical activity of the oil measurements. Oil showed a moderate antiradical activity, reducing quantity of produced hydroxyl radicals to about 20% of initial value. This study succeeds in creating directly comparable and quantitative data for the oil unsufficiently examined so far.

Njegova aktivnost na gljivu T. viride bila je jača od aktivnosti koju je pokazao antifungalni lek bifonazol upotrebljen kao pozitivna kontrola (MIK 100 i 200 μg/ml, redom). Ovaj rad ukazuje na činjenicu da R. ontariense predstavlja obećavajući izvor prirodnih proizvoda sa antifungalnom aktivnošću.

AB  - The antifungal activity of dimethyl sulfoxide (DMSO) extracts of three bryophyte species, two mosses (Atrichum undulatum (Hedw.) P. Beauv., Physcomitrella patens (Hedw.) Bruch and Schimp) and a liverwort (Marchantia polymorpha L. ssp. ruderalis Bischl. and Boisselier), grown in nature and in axenic culture was evaluated by microdilution method against five fungal species (Aspergillus versicolor, Aspergillus fumigatus, Penicillium funiculosum, Penicillium ochrochloron and Trichoderma viride). All the investigated bryophyte extracts have been proved to be active against all fungi tested. In general, extracts made from material grown in the laboratory (in vitro) conditions express better antifungal activity comparing to those made from material grown in the nature. Some of the fungi tested react similarly to both extracts.

All the investigated bryophyte extracts have been proved to be active against all fungi tested. In general, extracts made from material grown in the laboratory (in vitro) conditions express better antifungal activity comparing to those made from material grown in the nature. Some of the fungi tested react similarly to both extracts.",

AB  - Rapid micropropagation of Nepeta rtanjensis, N. sibirica and N. nervosa was performed. Qualitative and quantitative nepetalactone content in methanol extracts of in vitro grown plants was analysed by reverse-phase HPLC coupled with UV and MS detection. Only trans,cis-nepetalactone was detected in shoots of N. rtanjensis, while cis,trans-nepetalactone stereoisomer was present in N. sibirica. No nepetalactone was observed in shoots of N. nervosa. The antimicrobial activity of methanol extracts, against eight bacterial and eight fungal species, was evaluated. All the tested extracts showed significant antibacterial and strong antifungal activity. However, N. rtanjensis extract exhibited the best antimicrobial potential.

AB  - The aim of our study was to investigate the effect of camphor essential oil on rat cerebral cortex activity by fractal analysis. Fractal dimension (FD) values of the parietal electrocortical activity were calculated before and after intraperitoneal administration of camphor essential oil (450-675 mu l/kg) in anesthetized rats. Camphor oil induced seizure-like activity with single and multiple spiking of high amplitudes in the parietal electrocorticogram and occasional clonic limb convulsions. The FD values of cortical activity after camphor oil administration increased on the average. Only FD values of ictal ECoG sequences were lower than those before camphor oil administration.

AB  - We have decided to detect the seizure brain activity induced by camphor essential oil in rats, as there are still reports of severe intoxication in humans by camphor. The experiments were performed on adult Wistar male rats. The surgery and the acute electrophysiological recordings were done under pentobarbital sodium anesthesia. Local field potentials of cerebral parietal cortex were recorded. The rats were injected by camphor essential oil (150-675 μl/kg b.w., in 1 ml of saline, i.p). We analyzed by Fast Fourier Transform the digitized electrocortical signals before and up 2-3 h after essential oil administration at the sampling rate of 256 Hz. There was a great increase of the mean total power spectra, a relative increase of the power in delta range and decreases of theta and beta ranges when seizural activity with spikes and waves of large amplitude occurred at the level of cerebral cortex 5-30 min after administration of camphor oil at doses >300 μl/kg b.w. It is necessary to investigate further the dose dependent effectiveness of each terpenoid constituent of camphor essential oil on brain activity in order to elucidate the relative safety of the usage of essential oils with these terpenoids.
