AB  - Phenols are highly toxic organic compounds found in wastewater due to various industries’ pollution. Its removal is of great importance for human and animal health. Enzymatic wastewater treatment has several advantages over traditional methods. Enzyme immobilization onto solid carriers enables its reusability and lowers the cost of treatment. In this work, immobilized horseradish peroxidase on chemically modified alginate hydrogel was tested for phenol removal. The reusability of the tested immobilizate was monitored in repeated cycles. After five consecutive cycles, the remaining activity of the immobilized enzyme was 54%. The obtained result shows the potential for using this immobilizate for wastewater treatment.

AB  - In nature, woody substrates are usually degraded by entire communities of microorganisms, which are nowadays jeopardised by anthropogenic influence, making it important to define the functional specificity of every species. Two strains of autochthonous fungi from Serbian lowland forests (Pleurotus ostreatus Ser1 and Cyclocybe aegerita Ser1) have been investigated for their ability to degrade lignin substrates [oak sawdust, oak isolated cell walls, and synthetic dehydrogenative polymer (DHP)]. Measuring the activities of the enzymes involved in lignin degradation was coupled with detecting the HPLC profile of the phenolics in the fungal growth media, and the lignin loss. While Pleurotus ostreatus Ser1 appeared highly effective within a very short time span, Cyclocybe aegerita Ser1 failed to degrade lignin. This situation was supported by very high enzyme activities and the low presence of phenolics in the media of Pleurotus ostreatus Ser1, compared to very low enzyme activity and the high presence of phenolics in the media with Cyclocybe aegerita Ser1.

AB  - In this study, we investigated, in an in vitro system, the interaction of SiO2 (as a complex with NH4OH) with the peroxidase-catalyzed polymerization of lignin monomer into lignin model compound DHP, imitating conditions of the last step of lignin formation in the cell walls. The structure of obtained polymer was monitored using fluorescent spectroscopy and AFM 30 minutes after the start of synthesis. We studied effect of two different Si concentrations, 0.6 mM and 6 mM. Analysis of the DHP fluorescent spectra, and the application of mathematical decomposition clearly shows that structure of lignin model compound depends on Si concentration in the reaction mixture. The amount of Si affects the distribution of electrons in the DHP polymer. When silicon is present at a concentration of 6 mM, the position of the longest-wavelength APD band shifts towards shorter wavelengths compared to the position of the APD band for 0.6 mM Si. This indicates that during polymer synthesis, higher concentration of silicon may inhibit the formation of pi-electronic structures, which are responsible for electron delocalization. AFM images also show differences in the size and regularity of DHP globules.

AB  - Semena kukuruza (Zea mais L.) mogu biti kontaminirana sa jednim od najpasnijih prirodnih kancerogenih kontaminanata kao što su aflatoksini. U ovoj studiji, proučavan je uticaj aflateritoksina na strukturne karakteristike strukture. Korišćena je fluorescentna spektropija u kombinaciji sa matematičkom analizom spektra. Primenjen je matematički model za analizu spektra asimetričnog modela (Log-normal) koji koristi Nelder-Mead algoritam (Matlab 6.5). Rezultati razlaganja fluorescentnih spektara pokazuju četiri glavne fluorescentne fluorescentne emisije sa 054 emisiona približna 050 komponenta. Ovaj rezultat može da se koristi za praćenje promene u semenu usled prisustva aflatoksina. Glavni pokazatelj uticaja aflatoksina na strukturu kukuružnog semena je pomeraj dugo-talasne emisione komponente (520 nm).

DETERMINATION OF COUMAPHOS RESIDUES IN HONEY AFTER VARROA TREATMENT USING FLUORESCENCE SPECTROSCOPY

AB  - Varroa infestation is considered a major threat to honeybees due to its strong impact on the colony, attacking larval cells, increasing bee mortality, and serving as a vector of pathogens. Coumaphos is an organophosphate-based acaricide insecticide which is frequently used to control Varroa (Varroa destructor) mites in the honey bee (Apis melifera) colonies. The accumulation of these lipophilic contaminants in honey and beeswax may affect honey bee health, mainly honey bee larvae which are the most sensitive, and negatively affect queen quality. In this study, we analyzed honey samples from hives of honey bees that were treated with Coumaphos, as well as the samples of pure Coumafos. The front-face fluorescence measurements of analyed honey samples were recorded using an Fl3-221 P spectrofluorimeter, with a 450 W Xenon lamp. Obtained results showed an emission peak with maximum around 400 nm, after excitation at 340 nm. It could be related to the presence of Coumaphos in honey samples, which showed the characteristic position of emission maximum at the same wavelength. These results showed that fluorescence spectroscopy may be useful as a fast and sensitive method, for the efficient and specific determination of toxic substances such as Coumaphos residues in honey. Due to the proven toxic effects on honey bees, as well as on humans, monitoring hazardous contaminants in honey is of crucial importance in the field of food and agriculture.

Coumaphos is an organophosphate-based acaricide insecticide which is frequently used to control Varroa (Varroa destructor) mites in the honey bee (Apis melifera) colonies. The accumulation of these lipophilic contaminants in honey and beeswax may affect honey bee health, mainly honey bee larvae which are the most sensitive, and negatively affect queen quality. In this study, we analyzed honey samples from hives of honey bees that were treated with Coumaphos, as well as the samples of pure Coumafos. The front-face fluorescence measurements of analyed honey samples were recorded using an Fl3-221 P spectrofluorimeter, with a 450 W Xenon lamp. Obtained results showed an emission peak with maximum around 400 nm, after excitation at 340 nm. It could be related to the presence of Coumaphos in honey samples, which showed the characteristic position of emission maximum at the same wavelength. These results showed that fluorescence spectroscopy may be useful as a fast and sensitive method, for the efficient and specific determination of toxic substances such as Coumaphos residues in honey. Due to the proven toxic effects on honey bees, as well as on humans, monitoring hazardous contaminants in honey is of crucial importance in the field of food and agriculture.",

AB  - Silica is beneficial element for plants. Many studies of silica show the useful effects that Si can provide on plant-environment relationships in a wide variety of crops, as increase in growth and yield and improvement of resistance to metal toxicity, salt stress, drought resistance etc. The occurrence of Si within a plant is a result of its uptake in the form of soluble Si(OH)4 or Si(OH)3O ion from the soil, and its polymerization at a final location. Si is mostly accumulated in the cell walls (CWs), thus providing strengthening of CWs, increasing mechanical strength, and alleviating effects of various biotic and abiotic types of stress. Lignin, as one of the main components of the plant CW, is a natural phenolic polymer with high molecular weight and complex structure. Accumulation of silica in plants is directly connected with chemistry and structure of lignin. Silicon affects the composition of CWs by its binding to the phenolic and carbohydrate CW components in polymerization reactions by altering linkages of non-cellulosic polymers and lignin. We studied, in an in vitro system, the interaction of SiO2 with the peroxidase catalyzed polymerization of a lignin monomer into the lignin model compound (DHP). Fluorescence microscopy and spectroscopy combined with AFM technique showed that Si was bound to the final polymer, and the structure of the Si-DHP differed from pure DHP. We show that Si significantly influence on lignin structure. The results contribute to understanding of Si binding to lignin in cell walls, which may have impact on possible Si applications in agriculture.

AB  - Mung bean (Vigna Radiata L.) seeds are an important source of both nutrients (such as proteins, fibers, vitamins) and a variety of bioactive compounds (like phenolic compounds). Ultraviolet (UV) light has an important function as a major environmental signal important for plant growth and development, but at the same time it may cause certain damaging effects on macromolecules and other cellular components. Plants respond to the production of reactive oxygen species (ROS), caused by UV irradiation, by activating changes in morphology, physiology, or production of secondary metabolites. Some of these mechanisms increase antioxidant capacity in order to reduce the harmful effect of produced ROS. In the present research, the duration of exposure to UV-A irradiation and its influence on antioxidant activity of mung bean seeds were studied. Seeds were exposed to constant irradiation for 1 or 3 hours using a UV-A lamp (93μW/cm2) at a 50 cm distance. Antioxidant activity was tested using DPPH (2,2-Diphenyl-1-picrylhydrazyl) assay. The results indicate a significant increase in antioxidant activity of the UV-A irradiated seeds after 1 hour (74,45 % ± 0,40), compared to the control (72,85 % ± 1,55). After prolonged exposure, measured antioxidant activity significantly increased (77,99 % ± 0,71). Our results show a correlation between duration of UV-A irradiation and the increase in antioxidant activity. This could be beneficial in agriculture for producing fortified food.

AB  - Controlled drug delivery systems are of great interest in modern science and medicine. This study was oriented towards encapsulation of synthetic lignin (DHP) into alginate beads and investigation of its release kinetic. Encapsulation was achieved by the ionotropic gelation method. In vitro release profiles were fitted into four release kinetic models (zero-order, first-order, Higuchi, and Korsmeyer-Peppas). According to the in vitro release, data alginate beads showed initial burst release followed by prolonged release. The best correlation was found for the Korsmeyer-Peppas model (R2 = 0.948), with an n value lower than 0.43, indicating the DHP transport mechanism was controlled by quasi-Fickian diffusion. This delivery system could be promising for chronic wound care and antibacterial treatment.

AB  - Silicon (Si) is considered a beneficial element for plants, mostly accumulating in cell walls, where its location and content are primed by the chemistry and structure of lignin. It is unrevealed how Si interacts with the process of lignin formation in the CWs. We studied, in an in vitro system, the interaction of SiO2 with the peroxidase-catalyzed polymerization of a lignin monomer into the lignin model compound, imitating conditions of the last step of lignin formation. FTIR and fluorescence spectroscopy and microscopy showed that Si is bound to the final polymer, and the structure of the Si-DHP differs from pure DHP. Fluorescence spectroscopy showed that Si does not bind to the monomers, so Si probably inhibits the formation of the larger lignin fragments, as evidenced by HPLC-DAD, by binding to dimmers formed during DHP synthesis. The structural changes of the polymer are related to the changed proportion of the fractions of various MW. The enzyme catalyzing DHP synthesis was not inhibited by Si. HRP activity was increased in presence of Si except for 6 mM Si. This may indicate that the complex formed with Si and short oligomers activates the enzyme, and prevents the formation of the large fragments.

AB  - BACKGROUND Aflatoxin B-1 (AFB(1)) is the most dangerous of the mycotoxins that contaminate cereal seeds naturally. The relationship between AFB(1) and changes in lignin organic free radicals in seeds is not known, nor is the part of the seed that is more targeted. Using optical and electron paramagnetic resonance spectroscopy, we investigated AFB(1)-induced changes in lignin and organic free radicals in seeds, and whether the inner and outer seed fractions differ in response to increasing AFB(1). RESULTS Different changes in the content of lignin and free radicals with increasing AFB(1) concentrations were observed in the two seed fractions. There was a significant positive linear correlation (R = 0.9923, P = 0.00005) between lignin content and AFB(1) concentration in the outer fraction, and no correlation between the lignin content and the AFB(1) concentration in the inner fraction. We found a positive correlation between the area of the green spectral emission component (C4) and the AFB(1) concentration in the outer fraction. CONCLUSIONS To the best of our knowledge, the results showed, for the first time, that maize seed fractions respond differently to aflatoxin with regard to their lignin and organic free radical content. Lignin content and (C4) area may be reliable indicators for the screening of lignin changes against AFB(1) content in the seeds, and thus for seed protection capacity.

AB  - Understanding of composition and connections between the building macromolecules of plant
biomass, such as cellulose, hemicellulose and lignin, is main key for their better utilization in
biofuels industry. We compared four different plant species which are abundant in the region of
the Western Balkans. We investigated the structure of the cell walls, as the main constituent of
plant biomass, isolated from branches of softwood (Picea omorika (Pancic) Purkine), hardwood
(Acer platanoides L.), maize stem (Zea mays L.) as examples of crop species, and Paulownia
tomentosa tree as a fast-growing species with a huge biomass yield. For our investigation, we
combined Fluorescence-detected linear dichroism (FDLD) method and X-ray Diffraction. We
obtained data for anisotropy and crystallography which are a base for prediction of the best and
appropriate plant species for easy deconstruction of its biomass. Our results show that Acer
branch as a hardwood shows the highest anisotropy and the lowest crystallinity compared to the
other species while Picea Omorika needles show opposite results as the lowest anisotropy and
the higher crystallinity. The results for maize show that the stems are easier for utilization than
leaves. The isolated cell walls from leaves of Paulownia tomentosa show similar results and
good correlation between anisotropy and crystallinity, thus we can conclude that this plant is
easy to use in biofuel industries.

AB  - The effect of acetonitrile (ACN) on the micellization of a cationic surfactant, cetyltrimethylammo-nium bromide (CTAB), in aqueous solutions at different temperatures was studied. The critical micellar concentration (CMC) as well as the degree of counter ion dissociation (a) of CTAB in aqueous ACN mixtures (10-20 % v/v) at various temperatures (291.2-298.2 K) were determined by electrical conduc-tivity measurements, while steady-state fluorescence measurements were used to determine several CMCs (for comparison) and a micellar aggregation number (N-agg) as well. At a fixed temperature, both an increase in CMC and a decrease in N-agg were observed for an increase in ACN in the solvent mixture. With a temperature increase, CMC values increased for 10 % (v/v) ACN, while for both 15 % and 20 % (v/v) ACN, CMC values were a minimum at T = 295.2 K with a temperature increase. In addition, some aspects related to the Krafft temperature behavior of CTAB in the examined micellar systems are discussed.

Microstructural and mechanical characteristics of self-compacting concrete with waste rubber

AB  - Due to the increasing environmental pollution caused by scrap tires, a solution is being sought to recycle and use them in a field of civil engineering, i.e., construction. This paper will provide a brief overview of previous researches that give detailed information on the advantages and disadvantages, considering the microstructural and mechanical characteristics of self-compacting concrete, when waste tire rubber as an aggregate is added. With this aim, a database of 144 different mixtures of self-compacting concrete with partial substitute of natural aggregate with recycled tire rubber (self-compacting rubberized concrete, SCRC) provided by various researchers was created. In this study we show that Gaussian process regression (GPR) modelling is an appropriate method for predicting compressive strength of SCC with recycled tire rubber particles and is in accordance with the results displayed by SEM images.

AB  - Phenolic compounds are one of the most common pollutants in aqueous systems, so their removal
from water is of major interest. Among biocatalysts used for phenol removal, horseradish peroxidase
is the most investigated for this purpose. Enzyme inactivation is a major problem which could be
successfully overcome by immobilization of the enzyme onto different polymers. Tyramine-alginate
micro-beads were tested for the immobilization of horseradish peroxidase. Different concentrations of
tyramine-alginate were used and their influence on specific activity of the enzyme was tested.
Increasing concentration of oxidized alginate results in increase of specific activity. Immobilized HRP
was tested for phenol removal in a batch reactor. Presented results were obtained with HRP
immobilized within 10 mol% tyramine-alginate micro-beads. These biocatalysts can be used up to
three cycles.

AB  - Horseradish peroxidase (HRP, E.C. 1.11.1.7) catalyzes oxidation of aqueous aromatic compounds
using hydrogen peroxide. Enzymatic treatment methods for phenol removal from wastewaters has
become an efficient and environmentally friendly alternative for the traditional methods.
Carboxymethyl cellulose (CMC) derivative with tyramine attached via amide bond to carboxyl groups
has been chosen as carrier for immobilization. In effort to overcome the main disadvantage of
entrapment immobilization method, loss of enzyme activity due to washing out from the carrier, HRP
was modified in a reductive amination reaction and tyramine was bound to the enzyme.
Immobilization of tyramine-HRP onto tyramide-carboxymethyl cellulose carrier was carried in an
emulsion polymerization reaction that produced carboxymethyl cellulose microbeads. The highest
specific activity of the obtained biocatalyst was 0.227 U/ml and after 48h of storage 0.197 U/ml.
Immobilized tyramine-HRP retained 87% of activity after 48 h. Immobilized HRP is a suitable
candidate for wastewater treatment.

AB  - Cell wall can be considered as a nano-composite in which cellulose, lignin and hemicelluloses are
interconnected in a specific manner. Mechanical and physical propreties of plant fibres are dependent
on the orientation of constituent polymers (cellulose, hemicellulose, lignin). Fourier transform
infrared (FTIR) microscopy was used to examine the orientation of the main plant polymers in
transversal and longitudinal direction of the isolated cell wall of the maize leaves. Polarised FTIR
measurements indicated an anisotropy, i.e. orientation of the cellulose microfibrils that was more or
less parallel to the longitudinal axis of the cell wall. Xylan has parallel orientation with regard to the
orientation of cellulose, as well as lignin.

AB  - Steady state fluorescence spectroscopy in combination with Principal Component Analysis (PCA) for
spectral analysis was used to differentiate multifloral honeys from different parts of Serbia. The
emission spectra were recorded in the wavelength range 280 – 550 nm, after excitation in the 270 –
370 nm range. After normalization of the spectra, chemometric evaluation of the spectral data was
carried out using principal component analysis (PCA). This study indicates that front-face
fluorescence spectroscopy is a promising technique for the authentication of geographical origin of
honey and may also be useful for determination of the botanical origin within the same unifloral honey
type.

AB  - Plant cell walls are composed of a framework of cellulose microfibrils that are
interconnected with heteropolysaccharides (lignin, hemicelluloses) in a specific
manner. Plant cell walls form a large part of the plant body and define its
characteristics. Structural organisation of the cell wall and related polymers is
important for both mechanical properties of plants and chemical reactions occurring
in the wall space, especially in the response to stress.By using imaging FTIR
microscopy, run in transmission mode and at different polarisation modes (from 0° to
90°), it is possible to follow the chemical variability and the orientation of cell wall
polymers (cellulose, hemicelluloses and lignin) of the Arabidopsis thaliana stem. The
polarised FTIR measurements indicated that both xylan and lignin have parallel
orientation with regard to the orientation of cellulose. It is believed that this structuring
of lignin in the S2 layer of the cell wall might be a result of the spatial constraints
within the cell wall, occuring due to the previous deposition of
cellulose/hemicellulose in a strongly oriented assembly.

abstract = "Plant cell walls are composed of a framework of cellulose microfibrils that are
interconnected with heteropolysaccharides (lignin, hemicelluloses) in a specific
manner. Plant cell walls form a large part of the plant body and define its
characteristics. Structural organisation of the cell wall and related polymers is
important for both mechanical properties of plants and chemical reactions occurring
in the wall space, especially in the response to stress.By using imaging FTIR
microscopy, run in transmission mode and at different polarisation modes (from 0° to
90°), it is possible to follow the chemical variability and the orientation of cell wall
polymers (cellulose, hemicelluloses and lignin) of the Arabidopsis thaliana stem. The
polarised FTIR measurements indicated that both xylan and lignin have parallel
orientation with regard to the orientation of cellulose. It is believed that this structuring
of lignin in the S2 layer of the cell wall might be a result of the spatial constraints
within the cell wall, occuring due to the previous deposition of
cellulose/hemicellulose in a strongly oriented assembly.",

AB  - DP-LS microscopy is a useful and easily applicable technique for rapid screening
of cell wall structural order, by mapping the linear dichroism of cellulose fibrils.
It allows screening and quantification of the alignment of cellulose fibrils, relative
to one another, in cell walls. The goal was to provide new information about
structural characteristics of the isolated cell walls among species which may be a
basis for their possible applications, such as biofuel and biomaterial industry, but
also to understand what plant response to stress is.
The DP-LSM method is shown to be a simple and rapid way to check the structural
complexity in the cell wall through cellulose labeling.

AB  - Ćelijski zid vlakana drveta može se smatrati kao nano-kompozit, u kojem su celuloza,
lignin i hemiceluloze kompleksno povezani. Organizacija i svojstva polimera drveta u
velikoj meri određuju svojstva vlakana i drveta, pa je i razumevanje interakcija između
ovih polimera ključ za genetski razvoj poboljšanog drveta i kvaliteta vlakana.1
Polarizovana FTIR mikro-spektroskopija se pokazala kao korisno sredstvo u proceni
anizotropije, odnosno orijentacija polimera ćelijskog zida u strukturi vlakna.2 Orijentacija
celuloze, glukomanana, ksilana i lignina, kao suštinskih komponenti drveta, tako može
biti analizirana u odnosu na osu vlakana. Ispitivane su strukturne razlike između tvrdog i
mekog drveta na primeru grana javora (Acer platanoides L.) i Pančićeve omorike (Picea
omorika (Pančić) Purkyně). Pokazano je da su hemiceluloze orijentisane paralelno u
odnosu na vlakna celuloze, tj u odnosu na osu ćelijskog zida. Takođe, kao važan rezultat
je dobijeno da je lignin i kod javora i kod Pančićeve omorike paralelno orijentisan u
odnosu na osu, kao i na celulozu. I pored različitih prekursora lignina i različitih
hemiceluloza kod ove dve vrste drveta lignin ima sličnu orijentaciju.

AB  - Plant cell walls represent the most abundant, renewable and biodegradable composite on Earth. Its highly complex structure consists mainly of three organic compounds: cellulose, hemicelluloses, and lignin. Cell walls have wide applications in different industries, especially for biofuels and biomaterials. Fluorescence spectroscopy is the method allowing investigation of cell wall structure thought monitoring of lignin autoflorescence and thus interactions of lignin with the other cell wall constituents. Deconvolution of fluorescence spectra reveals the number and location of spectral component peaks by calculation of the approximation of the probability density (APD) of component positions. A characteristic of complex CW fluorescence is that the emission spectrum contains multiple log–normal components originating from different fluorophores, shorter wavelengths corresponding to phenolic structures and longer wavelengths to conjugated structures in lignin. Fluorescence spectroscopy has been used for fast screening of the cell wall properties from plants of different origin (hardwood, softwood and herbaceous plant), that may be important for selection of plants for possible applications. Fluorescence spectroscopy may be applicable in the investigation of the effect of stress on the cell wall. Lignin fluorescence emission spectra, peak intensities and shifts in the positions of the long-wavelength spectral components may be indicators of changes in cell wall structure during the stress. There is an increasing application of quantum dots (QDs) in plant science, as fluorescent markers. The isolated cell wall is an appropriate object for study of the interactions with nanoparticles. The results of different physico-chemical techniques including fluorescence spectroscopy combined with spectral deconvolution, show that in the cell walls, CdSe QDs predominantly bind to cellulose, via OH groups, and to lignin, via the conjugated C=C/C–C chains. Variability of bond types in lignin is related to the involvement of this polymer in plant response to various types of stress, by introducing local structural modifications in the cell wall. Different lignin model compounds have been used in order to reveal spectroscopic properties of lignin. Lignin model polymers were synthesized from three monomers, coniferyl alcohol, ferulic acid and p-coumaric acid mixed in various ratios, simulating lignin synthesis in the real cell walls. Further, by using fluorescence spectroscopy and appropriate mathematical methods, it is possible to get deeper insight into the structural characteristics of the molecule. Future investigations will be based on synthetic cell walls and on variation in a portion of all three main components: cellulose, hemicelluloses and lignin, also having in mind results of fine structural modifications in lignin model compounds.
