AB  - This study was focused on in vitro screening of the total antioxidant activity of the selected extracts of the mushroom Coprinus comatus and quinic acid, one of their antidiabetic ingredients, by an uncommon electrochemical assay. Indeed, direct current (DC) polarographic HydroxoPerhydroxo Mercury(II) Complex (HPMC) assay based on decrease of anodic limiting current originating from HPMC formation in alkaline solutions of hydrogen peroxide at potential of mercury dissolution, observed upon gradual addition of antioxidants, was applied herein for the estimation of the natural products' antioxidativity. Quinic acid was found to exhibit most promising antioxidant potential (4.0 +/- 0.2%mu L-1) being approximate to 2-fold more active than the screened C. comatus extract samples. Actually, such a finding puts some light on the antioxidativity of cyclic polyols, well understimated class of organic compounds, compared to aromatic (poly)phenolics. As a low cost, easy-to handle and accurate this polarographic assay may be thoroughly recommended for much broader use.

AB  - The phosphates (Pi) are nowadays recognized as pollutants. We studied the effect of Pi (0.625-12.500 mM KH2PO4) in the culture medium on in vitro grown 2-month-old Populus alba trees. The levels of sugar phosphates and vacuolar and cytoplasmic Pi in cell compartments of roots and stems were determined using P-31 NMR, while tissue-specific micro- and macroelements mapping on stem cross-sections were performed using synchrotron-based X-ray microfluorescence. Plants grown on 0.625 mM Pi (MS/2 medium) showed a survival rate of 70%. With the increase in Pi concentrations up to 6.250 mM, plant growth and survival increased, without changes in total P content per mass or in the levels of cytoplasmic and vacuolar phosphates, in both stems and roots, while the levels of Fe, Cu, Zn, Ca and Mn in stems increased. Further increase in Pi to 9.375 and 12.500 mM in the medium resulted in inhibited growth comparable with plants grown on MS/2, with the increase in total P content per mass up to 50%, in both stems and roots, but with no changes in cytoplasmic and vacuolar phosphates; 12.500 mM Pi affected even plant survival (70%) and thus might be considered as mildly toxic. P-31 NMR results indicate that the high tolerance of P. alba to increased Pi could result from its ability to maintain an intracellular P homeostasis, despite P accumulation up to 50%, in both stems and roots, indicating P. alba as a promising wood species for dendroremediation.

AB  - We present changes in Tacitus bellus antioxidative system that specifically correspond to subsequent phases of hemibiotroph Fusarium verticillioides infection revealed by histological analysis. T. bellus response to spore germination 6 h post inoculation (hpi), manifested as first oxidative burst, was characterized by transient decrease in malondialdehyde (MDA) content, transient increase in catalase (CAT), low level of superoxide dismutase (SOD) and peroxidase (POD) activity, as well as with transient decrease in total antioxidant capacity (TAC), total phenol content (TPC) and phenylalanine ammonium lyase activity (PAL), and no changes in polyphenol oxidase (PPO) activity, or phenolic profile. During the biotrophic phase of F. verticillioides infection, characterized by hyphae spread intercellularly in epidermal and mesophyll tissue, the host antioxidative system was suppressed. The transition to necrotrophic phase of F. verticillioides infection (inter- and intracellular colonization and sporulation), occurred 3-4 days post inoculation (dpi). During the necrotrophic phase, 5-7 dpi, slowed progression of colonization of T. bellus mesophyll cells occurred and it coincided with sharp increase in MDA content and CAT, SOD and POD activities, but the drop in TAC, TPC content, and PPO activity, as well as the production of phytotoxin fusaric acid. Presented results add to the knowledge of events and mechanisms related to the transition from biotrophy to necrotrophy in F. verticillioides.

AB  - BACKGROUND Cereal seeds, such as maize seeds, are frequently contaminated with aflatoxin B1 (AFB1), one of the most dangerous naturally occurring carcinogens. In plants, phenolamides are involved in biotic stress response. The data on variations of phenolamides in AFB1-containing seeds are limited. RESULTS Five polyamine conjugates, including two spermidine and three putrescine conjugates, were tentatively identified in methanolic extracts, using HPLC-DAD-MS. The ratio of putrescine to spermidine conjugates changed with increasing AFB1 concentration in a logistic dose-response manner, with a ratio of below 1 up to a concentration of 51.51 mu g kg(-1), and approximately 2.54 and 3 at higher concentrations of 177.4 and 308.13 mu g kg(-1), respectively. The observed variations of the total antioxidant activity and the total phenolic content may support this biphasic behaviour of the seeds against AFB1 stress. CONCLUSIONS The obtained data are a contribution to the understanding of the roles of polyamine conjugates in seed defence to increasing AFB1 concentrations. According to our knowledge, this study reports for the first time the biphasic response of maize seeds to increasing AFB1 contamination level, comprising the induction of polyamine conjugate accumulation and variation in the ratio of conjugates. This dose-response relationship may provide useful information in the field of agricultural and food chemistry as an indicator of AFB1 contamination level and, hence, for selecting an appropriate seed quality.

Phenolic Profiling of 12 Strawberry Cultivars Using Different Spectroscopic Methods

AB  - Phenolic compounds of 12 strawberry cultivars were profiled using spectrophotometry, spectrofluorometry, and high-performance liquid chromatography-mass spectrometry, coupled with multivariate regression analysis. Total phenolic content, total anthocyanin content, and total antioxidant capacity (TPC, TACY, and TAC, respectively) and concentrations of individual phenolics were evaluated, and the multivariate statistic was employed to identify the most promising cultivars based on their phenolic content. According to the principal component analysis, TAC was strongly correlated with the TPC (0.81), pointing out its importance in overall antioxidant activity. 'Joly', 'Laetitia', and 'Asia' cultivars were distinguished from others as the richest in concerning identified anthocyanins, almost all flavonoids and phenolic acids. Multivariate curve resolution with alternating least squares indicated the presence of two main types of fluorophores assigned to anthocyanins and phenolics, in which emission spectral ratios also showed the highest values in the referred cultivars. These cultivars could be recommended to consumers in terms of the health functionality of fruit.

Total phenolic content, total anthocyanin content, and total antioxidant capacity (TPC, TACY, and TAC, respectively) and concentrations of individual phenolics were evaluated, and the multivariate statistic was employed to identify the most promising cultivars based on their phenolic content. According to the principal component analysis, TAC was strongly correlated with the TPC (0.81), pointing out its importance in overall antioxidant activity. 'Joly', 'Laetitia', and 'Asia' cultivars were distinguished from others as the richest in concerning identified anthocyanins, almost all flavonoids and phenolic acids. Multivariate curve resolution with alternating least squares indicated the presence of two main types of fluorophores assigned to anthocyanins and phenolics, in which emission spectral ratios also showed the highest values in the referred cultivars. These cultivars could be recommended to consumers in terms of the health functionality of fruit.",

Pokazano je da BV formira stabilan koordinacioni kompleks sa jonima bakra u stehiometriji 1:1. Struktura BV je više planarna i energijski stabilna u kompleksu. Kompleks je pokazao jake paramagnetne efekte koji su pripisani nesparenom delokalizovanom elektronu. Delokalizovani elektron može poticati iz BV ili od jona Cu(II), pa je kompleks sastavljen ili od katjona BV radikala i jona Cu(I) ili od anjona BV radikala i jona Cu(III). Kompleks je podložan oksidaciji samo u prisustvu O2 i višku jona Cu(II), ili jakog oksidacionog sredstva, dok je otporan na redukciona sredstva. AB  - Biliverdin (BV) is one of the products of heme catabolism that is known to interact with redox-active metals. (Patho)physiological roles of BV and details of its interactions with metals under physiological conditions, which may be interconnected, are not fully understood. In the current work, we examined coordinate and redox interactions of BV with copper ions (Cu(II)). The interactions were exaimined under conditions that mimic physiological settings - in the phosphate buffer with pH 7.4. A battery of physico-chemical methods were applied, including UV-VIS spectrophotometry, fluorescence spectroscopy, nuclear magnetic resonance spectroscopy (NMR), mass spectrometry, Raman spectroscopy, electron paramagnetic resonance (EPR), and electrochemical methods. BV and copper formed a stable coordination complex with 1 : 1 stoichiometry, with BV showing a more planar structure and energetic stability in the complex. Antioxidant and antimicrobial activity of two Asplenium species

AB  - Extracts of the aerial (fronds) and underground parts (rhizome and roots) of Asplenium adiantum-nigrum L. and Asplenium ruta-muraria L. were analyzed for their phenolic composition and antioxidant properties (scavenging capacities against DPPH center dot and ABTS(center dot+)), and were comparatively assayed for antimicrobial and antibiofilm activity. Total of the 17 phenolic compounds, belonging to the classes of phenolic acids (hydroxybenzoic and hydroxycinnamic acids), flavonoids (flavan-3-ols, flavonols), and xanthones were identified and quantified using UHPLC/DAD/(-)HESI-MS/MS analysis. The present study shows that two analyzed ferns are significantly diverse in terms of their phenolic profile. Flavonoids considerably contribute to the total phenolic content of A. ruta-muraria, ranging from 47% in roots to around 75% in rhizome and fronds, while A. adiantum-nigrum preferably accumulates xanthones. Antioxidant activity of fern methanol extracts was ascribed to their phenolics content. Extracts of rhizomes of both Asplenium ferns exhibited the highest antimicrobial activity, while frond extracts were more efficient in inhibition of Candida albicans biofilm formation. The present study provides experimental evidence that methanol extracts from two analyzed Asplenium fern species exhibit strong antibacterial effect against Gram-negative bacteria which candidate them as a potentially useful natural antimicrobial agents.

AB  - In vitroacetylholinesterase (AChE) inhibitory activity of an autochthonous sample of the mushroomCoprinus comatus(encompassing fruiting body FB, mycelia M and filtrate F from the submerged cultivation) was the subject of this study.C. comatusF extract exhibited rather potent anti-AChE activity (73.0 +/- 1.5%) atin liquidconditions, comparable to those of the conventional drug donepezil (80.6 +/- 1.4%). Also, the same extract exhibited high anti-AChE activity (1 mu g)in solid. While its FTIR spectrum indicated the presence of phenolic compounds, quercetin (28.1 mu g g(-1)d.w.) was found to affect the observed bioactivity (59.8 +/- 0.9%). This is the first report of profound anti-AChE activity of anyC. comatusextract, a medicinal mushroom that has been successfully cultivated in P.R. China, due to the demanding needs of food industry.

AB  - Vanadium enters mycelium of fungus P. blakesleeanus in both +5 and +4 oxidation states. Mycelium in three stages of growth (20h, 36h and 56h) was treated with three concentrations of V+5 (1, 5 and 10 mM) for 1h or 5h. The decrease in viability of mycelium was noticed only at 36h for all applied concentrations. Glutathione has a role in reduction of V+5 to a less toxic form V+4 , so we assayed the changes in total glutathione. Mycelium in early exponential phase of growth (20 h), after 1h or 5h treatment, showed concentration dependent decrease in glutathione, with the largest decrease of 40±4% after 1h and 22±9% after 5h treatment induced by 10 mM V+5. In stationary phase of growth (56h), changes in glutathione concentration were noticed only after 1h of treatment, and the largest decrease (36±6%) was induced by 1 mM V+5. Activities of glutathione reductase (GR), glutathione transferase (GST) and glutathione peroxidase (GPx), have also been examined. Increase in the activities of GPx and GST was noticed in 56h old mycelia treated for 5h with 5 mM and 10 mM V+5, but the only statistically significant increase (54±20%) was noticed in GST activity after 5h of 10 mM V+5 treatment.

AB  - Reports about the influence of cerium-oxide nanoparticles (nCeO(2)) on plants are contradictory due to their positive and negative effects on plants. Surface modification may affect the interaction of nCeO(2) with the environment, and hence its availability to plants. In this study, the uncoated and glucose-, levan-, and pullulan-coated nCeO(2) were synthesized and characterized. The aim was to determine whether nontoxic carbohydrates alter the effect of nCeO(2) on the seed germination, plant growth, and metabolism of wheat and pea. We applied 200 mgL(-1) of nCeO(2) on plants during germination (Ger treatment) or three week-growth (Gro treatment) in hydroponics. The plant response to nCeO(2) was studied by measuring changes in Ce concentration, total antioxidative activity (TAA), total phenolic content (TPC), and phenolic profile. Our results generally revealed higher Ce concentration in plants after the treatment with coated nanoparticles compared to uncoated ones. Considering all obtained results, Ger treatment had a stronger impact on the later stages of plant development than Gro treatment. The Ger treatment had a stronger impact on TPC and plant elongation, whereas Gro treatment affected more TAA and phenolic profile. Among nanoparticles, levan-coated nCeO(2) had the strongest and positive impact on tested plants. Wheat showed higher sensitivity to all treatments.

AB  - Colored grains are rich sources of anthocyanins that could play an important role in the prevention of various diseases associated with oxidative stress. Bearing in mind that cereals are widely grown crops, anthocyanins-rich colored grains could be used as a functional food ingredient that provides health benefits to a large part of human world's population. This study investigated composition and content of anthocyanins in the grain of blue popping maize, deep purple maize, purple wheat, and black soybean. The relationship of acylated and non-acylated forms before and after alkaline hydrolysis of anthocyanin extracts has been studied as well. Deep purple maize had the highest content of total anthocyanins reaching as much as 4988.90 mg CGE/kg d.m. Ten anthocyanins were identified in blue popping maize, of which two are isomers of cyanidin-3-(malonylglucoside) and three of cyanidin-3-(dimalonyl-beta-glucoside). Seven, nine, and three anthocyanins have been identified in the deep purple maize, purple wheat, and black soybean, respectively. Cyanidin derivatives were predominant and their acylated forms accounted for about 98, 29, 71, and 0% of the total anthocyanins content in the grains, respectively. According to the study, acylated derivatives were completely degraded under the effect of highly alkaline pH. However, at the beginning of their degradation they were transformed to their non-acylated parents. The results could be useful to better understanding of the nature of anthocyanin in colored grains and, in that regard, their use for the derivation of food products with functional potential, as well as of natural dyes and pharmaceutical ingredients.

AB  - Active ingredient of the commercial herbicide BASTA (B), phosphinothricin, acts as an inhibitor of glutamine synthetase (GS), a key enzyme in ammonium assimilation. The treatment with BASTA leads to an elevation of ammonium levels in plants and further to various physiological alterations, ammonium toxicity and lethality. Results of the present study emphasize the complexity underlying control mechanisms that determine BASTA interaction with essential oil (EO) from Nepeta rtanjensis (NrEO), bioherbicide inducing oxidative stress in target plants. Simultaneous application of NrEO and BASTA, two agents showing differential mode of action, suspends BASTA-induced ammonium toxicity in Arabidopsis thaliana plants. This is achieved through maintaining GS activity, which sustains a sub-toxic and/or sub-lethal ammonium concentration in tissues. As revealed by the present study, regulation of GS activity, as influenced by BASTA and NrEO, occurs at transcriptional, posttranscriptional, and/or posttranslational levels. Two genes encoding cytosolic GS, GLN1;1 and GLN1;3, are highlighted as the main isozymes in Arabidopsis shoots contributing to NrEO-induced overcoming of BASTA-generated ammonium toxicity. The effects of NrEO might be ascribed to its major component nepetalactone, but the contribution of minor EO components should not be neglected. Although of fundamental significance, the results of the present study suggest possible low efficiency of BASTA in plantations of medicinal/aromatic plants such as Nepeta species. Furthermore, these results highlight the possibility of using NrEO as a bioherbicide in BASTA-treated crop fields to mitigate the effect of BASTA residues in contaminated soils.

AB  - Compared to plants, nowadays mushrooms attract more attention as functional foods, due to a number of advantages in manipulating them. This study aimed to screen the chemical composition (fatty acids and phenolics) and antioxidant potential (OH center dot, 2,2-diphenyl-1-picrylhydrazyl (DPPH center dot) and ferric reducing ability of plasma (FRAP)) of two edible mushrooms, Coprinus comatus and Coprinellus truncorum, collected from nature and submerged cultivation. Partial least square regression analysis has pointed out the importance of some fatty acids-more precisely, unsaturated fatty acids (UFAs) followed by fatty acids possessing both short (C6:0 and C8:0) and long (C23:0 and C24:0) saturated chains-and phenolic compounds (such as protocatechuic acid, daidzein, p-hydroxybenzoic acid, genistein and vanillic acid) for promising anti-OH center dot, FRAP and anti-DPPH center dot activities, respectively. However, other fatty acids (C16:0, C18:0 and C18:3n3) along with the flavonol isorhamnetin are actually suspected to negatively affect (by acting pro-oxidative) the aforementioned parameters, respectively. Taken together, design of new food supplements targeting oxidative stress might be predominantly based on the various UFAs combinations (C18:2n6, C20:1, C20:2, C20:4n6, C22:2, C22:1n9, etc.), particularly if OH center dot is suspected to play an important role.

AB  - Vanadijum je esencijalni mikroelement za mnoge žive organizme i učesnik je mnogobrojnih bioloških procesa kod njih. načajnost u ispitivanju ovog elementa porasla je naglo u poslednje tri decenije pretpostavkom o njegovom značajnom doprinosu u tretmanu raznih vrsta bolesti. Skoro sve potrebne informacije koje se tiču oksidacionog oblika, koordinacione geometrije, oligomernog stanja vanadijuma kao i kako prisustvo različitih substrata inhibitora enzimskih aktivnosti utiču na njegove karakteristike se mogu dobiti upotrebom i kombinacijom različitih spektroskopskih metoda. Ispitivanje promena strukture vanadijuma kao potencijalno toksičnog ali i metabolički korisnog elementa je našlo veliku primenu u ispitivanju njegovog metabolizma kod gljiva kao vrste organizama koji imaju sposobnost usvajanja i modulacije različitih vrsta elemenata. Različite vrste gljiva usvajaju vanadijum u različitim oblicima, metabolišu ga na različite načine i imaju različite odgovore na njegovo prisustvo. bog složenih fiziko-hemijskih karakteristika i osetljivosti na uslove u kojima se nalazi, ispitivanje vanadijuma zahteva istovremenu primenu više spektroskopskih tehnika. Primena NMR, EPR, Ramanske, IC, optičke i Rendgenske spektroskopije u izučavanju ovog elementa kod gljiva rasvetljava njegovu ulogu u živim sistemima i daje nam informacije o mogućnostima biotransformacije ovog elementa što može imati značajnu ulogu u mnogim granama nauke a pre svega u medicini i zaštiti životne sredine.

AB  - Laetiporus sulphureus (Bull.) Murrill 1920, (Ph. Basidiomycota, Cl. Agaricomycetes, O. Polyporales, Fam. Polyporaceae) je jestiva gljiva, široko rasprostranjena u Evropi, Aziji i Severnoj Americi. Izolovana micelija autohtone gljive (vrba obala Dunava, Novi Sad) kultivisana je na čvrstoj podlozi i u potopljenom medijumu. Cilj istraživanja bio je da se upotrebom NMR spektroskopije ispita uticaj vanadijuma na metabolizam fosfata. Utvrđeno je da signali pozicionirani oko 4 ppm i 2,6 ppm pripadaju šećernim fosfatima (SP) i da nakon tretmana sa 10 mM vanadatom dolazi do povećanja signala za 78%, odnosno 23%. Nakon ispiranja micelijuma dolazi do postepene transformacije jednog oblika u drugi, tako da posle sat vremena signal SP na 2,6 ppm postaje dominantan u odnosu na signal pozicioniran na 4 ppm. Promene signala SP mogu ukazati na efektivnost inhibitora ili stimulatora energetskog metabolizma. Pored SP, polifosfati (PolyP) prisutni u miceliji gljive, takođe imaju ulogu u skladištenju energije. Primećeno je da se nakon izlaganja nepovoljnjim uslovima signal PolyP smanjio, što je potvrđeno u studijama sa kvascima S. cereviseae1 i N. crassa2. Nasuprot tome, nakon tretmana vanadijumom kod L. sulphureus nije došlo do redukcije signala PolyP, već je zabeležen mali porast signala dugolančanih polifosfata što može ukazati na mehanizam detoksifikacije micelije L. sulphureus kao metaboličkog odgovora micelije na dodavanje vanadata.

AB  - Vanadijum je prelazni metal, koji se može naći u brojnim oksidacionim stanjima, pri čemu su najznačajnija +4 (vanadil) i +5 (vanadat). S obzirom da gljive apsorbuju vanadijum, one predstavljaju glavni put njegovog ulaska u ekosistem.1 Vanadat je izučavan kao potencijalni antidijabetski i antikancerogeni agens, ali zbog svoje komplikovane hemije se ne koristi u terapijama. Mnogi efekti vanadijuma na ćeliju nisu poznati, a među njima je i efekat na sisteme antioksidativne zaštite.2 U ovom istraživanju je ispitivana aktivnost superoksid dismutaza (SOD), peroksidaza (Prx) i katalaza (CAT) tokom različitih faza razvoja gljive Phycomyces blakesleeanus, a kao odgovor na tretman micelijuma vanadatom. Efekat je praćen kod gljiva starosti 20 h, 36 h i 56 h, koje su izlagane 10 mM vanadatu tokom 1 h, 3 h ili 5 h. U eksponencijalnoj fazi razvoja (20 h) zabeležen je porast aktivnosti Prx i to 45±7% nakon 1 h, odnosno 19±7% nakon 5 h tretmana. Na prelasku iz eksponencijalne u stacionarnu fazu razvoja (36 h) 10 mM vanadat nije doveo do promene aktivnosti Prx u tretiranim uzorcima, dok je u stacionarnoj fazi razvoja (56 h) ponovo zabeležen porast aktivnosti Prx i to 22±4% nakon 1 h, odnosno 31±19% nakon 3 h tretmana. Porast aktivnosti SOD je zabeležen samo u stacionarnoj fazi razvoja, nakon 1 h tretmana i iznosi 26±2%. Nije uočena jasna promena aktivnosti CAT tretiranih u odnosu na kontrolne uzorke

AB  - Elementarni selen je najmanje biotoksičan oblik selena u prirodi, dok selen-oksianjoni mogu biti toksični zbog značajne pokretljivosti i rastvorljivosti u vodi. Mikrobiološka redukcija selena +4 (selenit) u elementarni selen je stoga od presudnog značaja za smanjenje bioraspoloživosti ovog elementa.1 Visoka reaktivnost selen-oksianjona sa tiolnim grupama, kao i formiranje kiseoničnih radikala u reakciji sa glutationom implicira da oksidativni stres doprinosi toksičnosti selena.2 Tokom biološke redukcije selenita se proizvodi velika količina peroksida čime se indukuje ekspresija gena koji kodiraju enzime antioksidativnog metabolizma, što uključuje i enzime metabolizma glutationa.1 Micelijum gljive Phycomyces blakesleeanus star 24 sata je tretiran 10 mM rastvorom selenita. Uzorci su nakon određenih vremenskih intervala (1, 5, 10, 30, 60 minuta, 24 i 96 sati) ispirani i homogenizovani, nakon čega je meren sadržaj glutationa i specifična aktivnost enzima: peroksidaza (POD), katalaza (CAT), superoksid dismutaza (SOD), glutation peroksidaze (GPx), glutation-S-transferaze (GST) i glutation reduktaze (GR). Nakon dodavanja selenita utvrđen je pad količine ukupnog glutationa u micelijumu. Aktivnost POD i SOD je rasla do 60 minuta, nakon čega je opadala. Aktivnost CAT opadala je odmah po dodatku selenita. Najizraženija je bila promena aktivnosti GPx gde je zabeležen značajan porast, dok su promene aktivnosti GST i GR bile manje izražene

AB  - Vanadate is proposed to play a pivotal role in application of edible fungus Coprinus comatus for medical purposes. In this study the concentration of extracellular vanadate acceptable for the submerged cultivation of C. comatus mycelium was established. The mycelium could grow, and overcome vanadate toxic effects, up to the concentration of 3.3 mM. Moreover, in this condition, at the end of the exponential phase of growth, biomass yield was almost identical to that in the control. P-31 NMR spectroscopy showed that addition of 10 mM vanadate to the mycelium in the exponential phase of growth provoked instantaneous increase of a sugar phosphates level which could be related to changes in activities of glycolytic enzymes. Exposure to higher vanadate concentration was toxic for the cell. V-51 NMR measurements revealed that monomer of vanadate is present in the cytoplasm causing the metabolic changes. C. comatus has also capacity for vanadate reduction, as shown by EPR measurements, but vanadyl uptake is significantly less comparing to vanadate.

In this study the concentration of extracellular vanadate acceptable for the submerged cultivation of C. comatus mycelium was established. The mycelium could grow, and overcome vanadate toxic effects, up to the concentration of 3.3 mM. Moreover, in this condition, at the end of the exponential phase of growth, biomass yield was almost identical to that in the control. P-31 NMR spectroscopy showed that addition of 10 mM vanadate to the mycelium in the exponential phase of growth provoked instantaneous increase of a sugar phosphates level which could be related to changes in activities of glycolytic enzymes. Exposure to higher vanadate concentration was toxic for the cell. V-51 NMR measurements revealed that monomer of vanadate is present in the cytoplasm causing the metabolic changes. C. comatus has also capacity for vanadate reduction, as shown by EPR measurements, but vanadyl uptake is significantly less comparing to vanadate.",

in human nutrition. As minimally processed food product, it is an important source of vitamins, 
minerals and various antioxidative compounds. In our greenhouse experiment, two Salanova let tuce cultivars (‘Aquino‘ RZ- green and ‘Gaugin‘ RZ- red multi-leaf butterhead) were grown in the 
fertile soil during three successive seasons (autumn, winter and spring). The aim of the study was 
to examine the effect of different microbiological fertilizers (EM Aktiv, Vital Tricho and combination 
of EM Aktiv and Vital Tricho), cultivars and growing seasons on the total phenolic content and 
peroxidase activity. Our results showed that in all seasons red cultivar ‘Gaugin‘ had higher total 
phenolic content compared to green cultivar ‘Aquino‘. Their highest level was measured in au tumn with fertilizer Vital Tricho (549.14 µg GAE g-1 FW). The elevated phenolic status of red versus 
green cultivar was further strengthened by its higher level of peroxidase with the highest activity 
measured in spring with combination of fertilizers EM Aktiv and Vital Tricho (0.43 U mg-1 prot). The 
higher peroxidase activity and content of UV-absorbing phenolics, as plant-borne substrates of 
these enzymes, in the red cultivars indicate that the red leaves are better equipped to combat 
oxidative stress. Consequently, red cultivar ‘Gaugin‘, with increased synthesis of health-promoting 
antioxidants, could be considered as functional food in human diet.

AB  - In this work, two non-invasive and independent methods for the
characterization of aflatoxin-stressed wheat seeds were applied.Aflatoxins
(AFs) are secondary toxic fungi metabolites, which exhibit hazardous effects
on plants (phytotoxic), as well as on humans and animals. First, for 2D EPR
imaging, we used 3-CP spin probe for the estimation of changes in the redox
state of the aflatoxin-stressed seed. On the other side, an excitation-emission
fluorescence approach was used to evaluate the spectral profiles of the key
biological molecules. We determined their areas and the ratio of the two
emission peaks at 355 nm (Ex 290) and 430 nm (Ex 350), corresponding to
protein and phenol emission, respectively. Our results showed that the ratio
of the two peaks (p/f) decreased in the presence of AFs. Both methods
revealed to be useful for the rapid, non-invasive assessment of the seeds'
stress state induced by AFs.

Phenolic profiles of June bearing and everbearing strawberries

AB  - The delicious fruits of strawberry (Fragaria x ananassa Duch.), consumed in both fresh and pro­ cessed form, аге ап important source of health promoting  compounds.  Amongst  them, straw  ber ­ ries possess consideraЫe quantities of various phenolics. For this study, twelve strawberry cultivars,  9 June bearing ('Roxana; 'Arosa; 'Joly', 'Asia', 'АIЬа', 'Jeny', 'Laetitia; 'Garda; 'Premy'), and 3 ever-bearing types ('AIЬion','Capri' and'lrma'), were evaluated for anthocyanin, phenolic acid and ftavonoid con­  tent using  HPLC-MS.  Dominant  anthocyanin  form  in  all  cultivars  was  pelargonidin-3-glucoside with the highest values detected in 'Asia' and 'Joly' cultivars (487 and 464 µg -g 1 FW, respect ively). 'Joly' also stands out for acylated pelargonidin -3- gluco side and prunin content, since the highest values of pelargonidin-3-rutinoside and  ellagic  acid  were  detected  in 'Laetitia'  (33  and  43  µg -g 1 FW, resp ectively). Uppermost оп the ftavonoid list was 'Asia' which was the richest in p-coumaroyl hexose and its derivate (87 and 64 µg -g 1 FW, respectively), followed by'Joly' dominating in kaempferol 3-glucuronide and kaempferol 3-coumaroyl glucuronide .The identification of phenolic com­ pounds revealed some interesting differences correlated to  the  culti var:  June  bearing  cultivars 'Asia' and 'Joly' were distinguished from others as the richest in terms of identified phenolic com ­ pounds. Branding  the  cultivars  with  optimal  phenolic  composition  empowers  their  consumption for  human  health  and wellЬeing.

AB  - Efforts to breed salt tolerant crops could benefit from investigating previously unexplored traits. One of them is a tissue succulency. In this work, we have undertaken an electrophysiological and biochemical comparison of properties of mesophyll and storage parenchyma leaf tissues of a succulent halophyte species Carpobrotus rosii ("pigface"). We show that storage parenchyma cells of C. rossii act as Na+ sink and possessed both higher Na+ sequestration (298 vs. 215 mM NaCl in mesophyll) and better K+ retention ability. The latter traits was determined by the higher rate of H+-ATPase operation and higher nonenzymatic antioxidant activity in this tissue. Na+ uptake in both tissues was insensitive to either Gd3+ or elevated Ca2+ ruling out involvement of nonselective cation channels as a major path for Na+ entry. Patch-clamp experiments have revealed that Caprobrotus plants were capable to downregulate activity of fast vacuolar channels when exposed to saline environment; this ability was higher in the storage parenchyma cells compared with mesophyll. Also, storage parenchyma cells have constitutively lower number of open slow vacuolar channels, whereas in mesophyll, this suppression was inducible by salt. Taken together, these results provide a mechanistic basis for efficient Na+ sequestration in the succulent leaf tissues.

AB  - Lignins and lignin-derived compounds are known to have antibacterial properties. The wound healing agents in the form of dressings produce faster skin repair and decrease pain in patients. In order to create an efficient antimicrobial agent in the form of dressing in the treatment of chronic wounds, a composite hydrogel of bacterial cellulose (BC) and dehydrogenative polymer of coniferyl alcohol (DHP), BC-DHP, was designed. Novel composite showed inhibitory or bactericidal effects against selected pathogenic bacteria, including clinically isolated ones. The highest release rate of DHP was in the first hour, while after 24 h there was still slow release of small amounts of DHP from BC-DHP during 72 h monitoring. High-performance liquid chromatography coupled with mass-spectrometry showed that BC-DHP releases DHP oligomers, which are proposed to be antimicrobially active DHP fractions. Scanning electron microscopy and atomic force microscopy micrographs proved a dose-dependent interaction of DHP with BC, which resulted in a decrease of the pore number and size in the cellulose membrane. The Fourier-transform infrared absorption spectra of the BC-DHP showed that DHP was partly bound to the BC matrix. The swelling and crystallinity degree were dose-dependent. All obtained results confirmed BC-DHP composite as a promising hydrogel for wounds healing.

The effect of auxin (indole-3-acetic acid) on the growth rate and tropism of the sporangiophore of Phycomyces blakesleeanus and identification of auxin-related genes

AB  - The roles of fungal auxins in the regulation of elongation growth, photo-, and gravitropism are completely unknown. We analyzed the effects of exogenous IAA (indole-3-acetic acid), various synthetic auxins including 1-NAA (1-naphthaleneacctic acid) and 2,4-D (2,4-dichlorophenoxyacetic acid), and the auxin transport inhibitor NPA (N-1-naphtylphtalamic acid) on the growth rate and bending of the unicellular sporangiophore of the zygomycete fungus. Phycomyces blakesleeanus. Sporangiophores that were submerged in an aqueous buffer responded to IAA with a sustained enhancement of the growth rate, while 1-NAA, 2,4-D. and NPA elicited an inhibition. In contrast, sporangiophores kept in air responded to IAA with a 20 to 40% decrease of the growth rate, while 1-NAA and NPA elicited an enhancement. The unilateral and local application of IAA in the growing zone of the sporangiophore elicited in 30 min a moderate negative tropic bending in wild type C2 and mutant C148madC, which was, however, partially masked by a concomitant avoidance response caused by the aqueous buffer. Auxin transport-related genes ubiquitous in plants were found in a BLAST search of the Phycomyces genome. They included members of the AUX1 (auxin influx carrier protein 1), PILS (PIN-LIKES, auxin transport facilitator protein), and ABCB (plant ATP-binding cassette transporter B) families while members of the PIN family were absent. Our observations imply that IAA represents an intrinsic element of the sensory transduction of Phycomyces and that its mode of action must very likely differ in several respects from that operating in plant.

Effects of homocysteine and its related compounds on oxygen consumption of the rat heart tissue homogenate: the role of different gasotransmitters

AB  - The objective of this study was to investigate in vitro effects of 10 A mu M dl-homocysteine (dl-Hcy), dl-homocysteine thiolactone-hydrochloride (dl-Hcy TLHC), and l-homocysteine thiolactone-hydrochloride (l-Hcy TLHC) on the oxygen consumption of rat heart tissue homogenate, as well as the involvement of the gasotransmitters NO, H2S and CO in the effects of the most toxic homocysteine compound, dl-Hcy TLHC. The possible contribution of the gasotransmitters in these effects was estimated by using the appropriate inhibitors of their synthesis (N (omega)-nitro-l-arginine methyl ester (l-NAME), dl-propargylglycine (dl-PAG), and zinc protoporphyrin IX (ZnPPR IX), respectively). The oxygen consumption of rat heart tissue homogenate was measured by Clark/type oxygen electrode in the absence and presence of the investigated compounds. All three homocysteine-based compounds caused a similar decrease in the oxygen consumption rate compared to control: 15.19 +/- 4.01%, 12.42 +/- 1.01%, and 16.43 +/- 4.52% for dl-Hcy, dl-Hcy TLHC, or l-Hcy TLHC, respectively. All applied inhibitors of gasotransmitter synthesis also decreased the oxygen consumption rate of tissue homogenate related to control: 13.53 +/- 1.35% for l-NAME (30 A mu M), 5.32 +/- 1.23% for dl-PAG (10 A mu M), and 5.56 +/- 1.39% for ZnPPR IX (10 A mu M). Simultaneous effect of l-NAME (30 A mu M) or ZnPPR IX (10 A mu M) with dl-Hcy TLHC (10 A mu M) caused a larger decrease of oxygen consumption compared to each of the substances individually. However, when dl-PAG (10 A mu M) was applied together with dl-Hcy TLHC (10 A mu M), it attenuated the effect of dl-Hcy TLHC from 12.42 +/- 1.01 to 9.22 +/- 1.58%. In conclusion, cardiotoxicity induced by Hcy-related compounds, which was shown in our previous research, could result from the inhibition of the oxygen consumption, and might be mediated by the certain gasotransmitters.
