VARIJABILNOST MORFOLOŠKIH KARAKTERISTIKA LISTOVA DIVLJE KRUŠKE (Pyrus pyraster (L.) Burgsd.) NA PODRUČJU PIO „KOSMAJ“

AB  - Divlja kruška je na prostoru Srbije autohtona vrsta drveća koja je svrstana u
kategoriju vrsta „pod rizikom“. Hibridizacija sa kultivarima koji se uzgajaju za
proizvodnju voća je veoma česta, što otežava identifikaciju čistih divljih voćkarica i predstavlja jedan od ugrožavajućih faktora koji dovode do genetičke erozije
autohtonog genofonda. Cilj rada je utvrđivanje unutarpopulacione varijabilnosti
selekcionisanih test stabala divlje kruške na području Predela izuzetnih odlika
„Kosmaj“, kao osnove za konzervaciju raspoloživog genofonda. Listovi su sakupljeni
sa 16 test stabala selekcionisanih na području Kosmaja (100 listova po stablu), na
kojima su analizirani sledeći kvantitativni i kvalitativni parametri: dužina lisne
ploče, maksimalna širina lista, dužina lisne ploče od osnove lista do dela na kome
je merena maksimalna širina, dužina peteljke, ugao vrha lista, ugao osnove lista,
površina lisne ploče, obim lisne ploče, odnosno oblik lista, oblik osnove lista,
oblik vrha lista, obod lista. Sakupljeni listovi (ukupno 1600) su herbarizovani,
skenirani i izmereni pomoću softvera ImageJ, dok je statistička obrada podataka
urađena pomoću softvera Statgraphics Centurion XVII i Statistica 6.0. Materinsko
stablo 35 se izdavaja u odnosu na druga na osnovu dobijenih prosečnih vrednosti
kvantitativnih parametara, dok se materinsko stablo 27 izdvaja na osnovu kvalitativnih parameter. Rezultati istraživanja pokazali su da su sve dobijene vrednosti
statistički značajne (p < 0,05), što ukazuje na postojanje zadovoljavajućeg stepena
genetičke varijabilnosti između ispitivanih stabala, koji predstavlja dobru osnovu
za očuvanje i usmereno korišćenje genofonda ove vrste.

AB  - Poljski brest (Ulmus minor Mill.) je vrsta koja u šumskom fondu Republike
Srbije spada u kategoriju retkih/ugroženih vrsta čija je brojnost znatno smanjena
kao posledica holandske bolesti koja je prisutna na području čitave Evrope poslednjih decenija. Smanjenjem autohtone populacije poljskog bresta, šumarstvo je
lišeno njegovih dragocenih ekonomskih i meliorativnih vrednosti, a pejzažna
arhitektura uskraćena za upotrebu poljskih brestova u oplemenjivanju urbanih
sredina i vangradskih predela. Na prostoru predela izuzetnih odlika (PIO) „Kosmaj“
poljski brest je autohtona vrsta, čija je brojnost znatno redukovana, a populacija
svedena na nešto manje od 100 stabala kod kojih, u dužem vremenskom periodu,
nije konstatovan urod. Procenjena starost stabala, njihovo zdravstveno stanje,
nedostatak prirodnog podmlađivanja i plodonošenja kao i veliki broj faktora koji
dovode do genetičke erozije raspoloživog genofonda upućuju na potrebu namenske
proizvodnje sadnog materijala vegetativnim putem. U tom cilju obavljena je selekcija 12 materinskih stabala, na osnovu fenotipskih karakteristika i zdrvastvenog
stanja, sa kojih su sakupljene grančice od kojih su formirane reznice. U staklari
Šumarskog fakulteta, osnovan je klonski test potomstva u kome je tokom tri meseca praćen procenat prijema i preživljavanja reznica kao i uticaj debljine reznica
na njihovo ožiljavanje. Procenat preživljavanja se na kraju istraživanja kretao od
0,00% do 49,12% u zavisnosti od materinskog stabla, a prosečan prečnik na sredini
reznice za sva materinska stable iznosio je 3,58 mm. Dobijeni rezultati poslužiće
kao osnova za masovnu proizvodnju sadnog materijala za potrebe konzervacije i
održivog korišćenja genofonda poljskog bresta na ovom području.

AB  - Pedunculate oak (Quercus robur L.) is one of the economically and ecologically most
important deciduous forest tree species in Europe. In recent decades, there has been an
evident decline of pedunculate oak forests, caused by various factors, such as climate change,
site conditions, over-exploitation, or insufficient and inadequate regeneration. In Serbia, this
species usually occurs in the valleys of major rivers (Danube, Sava, and Morava), and the most
valuable pedunculate oak forests grow along the river Sava in the area of Srem, where there
are individual specimens aged hundreds of years. The protected area „Košutnjak Forest“
(Belgrade, Serbia), occupies 267 ha, with numerous forest tree species, among which are five
native oak species, including the pedunculate oak. Oaks can be categorized as „at-risk” species
in „Kosutnjak Forest”, since their populations are mostly represented by very old trees, with a
poor possibility of natural rejuvenation. This research aimed to determine the genetic
variability of the pedunculate oak population in the „Košutnjak Forest” using nuclear
microsatellites. DNA extraction was performed from young leaves, collected from 56 adult
trees, using a commercial peqGOLD Plant DNA Mini Kit (PEQLAB). In total 13 nuclear
microsatellites were used, and values of standard genetic diversity parameters were
calculated using GenAlEx 6.5 software. The number of alleles per locus ranged from 8 (MSQ13
and PIE239) to 39 (QrZAG90), with an average of 17.538. The number of effective alleles per
locus was in the range from 2,498 (QrZAG108) to 23.668 (QrZAG90), with an average of 8.360.
The average value of the observed heterozygosity (Ho) was 0.705, and the average unbiased
expected heterozygosity (uHe) was 0.818. A statistically significant deviation of the expected
heterozygosity from the observed heterozygosity occurs at 5 loci (MAQ4, PIE239, QpZAG104,
QrZAG108, and MSQ13). The average values of the fixation index were positive and
statistically significant, indicating an excess of homozygotes. According to the assessment of
the genetic status, determining the level of genetic variability of pedunculate oak served as a
basis for defining in situ conservation measures for the available gene pool in „Kosutnjak
Forest”.

AB  - The field elm (Ulmus minor Mill.) is an autochthonous tree species in Serbia that occurs as a
secondary species in oak forests. In the forest fund of Serbia, this species is rare/endangered
due to the genetic erosion and disappearance of elms from natural populations, mainly as a
result of Dutch elm disease. In the protected natural area „Kosmaj“ the field elm was identified
as one of the target species from the aspect of conservation. This research aimed to identify and
assess the state of available field elm genepool in the protected natural area „Kosmaj“, as a
basis for its conservation and sustainable use. By terrain reconnaissance a field elm gene pool
was identified, representing almost 100 trees: 47 individuals and 8 groups of trees, on an area of
652.99 ha. All trees were georeferenced and mapped. The average height of the trees is 10.34
m, the diameter at breast height is 15.68 cm, the perimeter is 51.09 cm, while the average
crown span is 3.95 m. Dry tops or dry side branches of trees, absence of yield for a long period,
and weak natural rejuvenation were recorded. The selected trees will be included
inconservation units, which were proposed to conserve the available gene pool in situ. To
conserve the available genepool ex situ, the cuttings from selected mother trees were collected
and the clonal test was established in the greenhouse of the Faculty of Forestry, University of
Belgrade. This clonal test could be a basis for the establishment of a field gene bank in the
future. It is planned to determine the variability of the selected trees, using morphological and
molecular markers. The initiated activities should contribute to the conservation and
sustainable use of the field elm gene pool in the protected natural area „Kosmaj“ and further
breeding of the species.

AB  - In situ conservation in Serbia is mainly presented in protected areas, such as national parks.
National Park „Kopaonik” is one of five national parks in Serbia, which covers a total area of
11,969.04 ha, including 13 localities with a protection regime of the first degree. Forest genetic
resources in the National Park „Kopaonik“ are affected by various biotic and abiotic threats
which lead to their genetic erosion. This research aimed to identify rare, endangered, relict,
endemic, and vulnerable forest trees at the locality „Duboka”, to assess their degree of
endangerment and propose conservation measures. The locality „Duboka” covers an area of
144.03 ha, at an altitude of between 1020 and 1780 m. By terrain reconnaissance, a total of 18
forest tree species, and 27 species in the shrub layer were identified, which indicates a
significant biodiversity of this locality. Nine forest tree species belong to the categories of rare
(rowan and wych elm), rare-endangered (silver birch), relict (European hop-hornbeam),
endemic (Balkan maple), and vulnerable species (aspen, wild cherry, wild pear, and whitebeam)
in Serbia. Among those species, it is very important to conserve the available gene pool of
European hop-hornbeam (Ostrya carpinifolia Scop.), which is a very rare, relict species that
occurs only in the locality „Duboka” in the entire area of Kopaonik, and the Balkan maple (Acer
heldreichii Orph. ex Boiss.), an endemic species whose range is drastically decreasing on the
entire Balkan peninsula. The measures of conservation and monitoring of individual genotypes
or groups of trees have been defined, and an assessment of the variability of gene pool using
genetic markers was proposed. In situ conservation units will include sites where all nine rare,
endangered, relict, endemic, and vulnerable forest tree species were selected and
georeferenced. The implementation of the proposed measures is expected in the following
period.

AB  - European White Elm (Ulmus laevis Pall.) is considered a rare and endangered
species in the forest fund of the Republic of Serbia. One of its native populations in Serbia
is conserved in the natural protected area „Veliko ratno ostrvo” (Belgrade). This research
aimed to assess the genetic and ecological potential of European White Elm from the
protected area „Veliko ratno ostrvo“ based on research conducted in a nursery progeny test.
The selection of test trees was done in the area of “Veliko ratno ostrvo”based on the yield
in spring 2019. The progeny test was established in the nursery of the Faculty of Forestry,
University of Belgrade. The assessment of gen-ecological potential was based on the survival
of the one- and two-year-old seedlings of 6 half-sib lines during two vegetations and their
morphological characteristics (height and root collar diameter). Statistical analyses were
performed with Statgraphics Centurion XVI software. The results of descriptive statistics,
one-way analysis of variance, and the least significant difference test were presented. The
percentage of seedlings' survival at the end of the second vegetation period was above 50%
in all halfsib lines. The differences in the measured characteristics were significant among
all half-sib lines (p<0.05), which indicates a strong effect of genetic control. The highest
mean value of the height and root collar diameter of one- and two-year-old seedlings was
recorded in half-sib line 4. The analysis of the gene-ecological potential of different European
White Elm half-sib lines will be continued in the pilot stand, established on the „Veliko ratno
ostrvo“, which will be used to monitor the survival and development of seedlings over a
longer period. Collected information on the gene-ecological potential of certain half-sib lines
will serve as a basis for further breeding of the species, through the selection of superior
genotypes.

AB  - Black poplar (Populus nigra L.) is one of the endangered native forest species in Europe, which is also endangered in
Serbia, with rare natural forests. One of the native black poplar populations in Serbia is conserved in the natural
protected area of „Veliko ratno ostrvo” Island, where this research was conducted. The wetland „Veliko ratno ostrvo”
Island is located in Belgrade (Serbia), at the confluence of the Sava and Danube rivers. One of the important
endangering factors for vegetation in this area is an irregular change in water levels. This research aimed to determine
the survival rate of two-year old black poplar seedlings after the period of flooding on the „Veliko ratno ostrvo” Island.
The seedlings were produced in March 2017, from cuttings that originated from 9 trees, previously selected in this area.
The seedlings were planted on the field in autumn 2018. This research was conducted in 2019, when the period of
flooding and water stagnation on the „Veliko ratno ostrvo” Island lasted 45 days (from middle May to the end of June),
with the highest water level at the beginning of June (the Danube water level was 603 cm, while the first flood alert is at
500 cm). The survival rate of seedlings was obtained based on the number of planted and the number of survived
seedlings after flooding. Seedlings originating from tree number 9 (clone number 9) had the highest survival rate after
the flooding period (85.70%), while the lowest value was recorded in clone number 8 (55.00%). More than 50% of
seedlings of all nine clones survived after the flooding. It can be stated that the seedlings of all nine clones have the
potential to be planted on the wetlands in Serbia, but this research should be continued with the aim of selecting
superior clones.

AB  - The main long-term goal of conservation and sustainable development of National
Park (NP) „Kopaonik” is to conserve and improve the natural values, including forest
genetic resources. It is located in central Serbia, covering an area of 11,969.04 ha.
One of the 13 localities with the first protection regime is „Jelak” (60.39 ha, 1000-1490
m a.s.l.). This research aimed to identify rare, endangered, relict, endemic, or species
„at-risk” at the locality „Jelak”, as a basis for proposing a program for its conservation.
Its main value is a presence of a mixed forest of yew and fir. By terrain reconnaissance,
a total of 13 forest tree species were recorded, among which Ulmus glabra (rare),
Betula pendula (rare/endangered), Populus tremula („at-risk“), and Taxus baccata
(relict), which indicates a significant diversity. In addition, 14 species were recorded in
the shrub layer, among which Acer heldreichii (endemic) and Acer platanoides (rare/
endangered). English yew (Taxus baccata L.) is a very rare species that occurs only in
the locality „Jelak” in the entire area of NP „Kopaonik”. This species is under varying
degrees of nature conservation in different European countries, and it is categorized
on the European Red List Category and Criteria („least concerning”). Since it is considered a tertiary relict in the forest fund of Serbia, it is important to conserve the available gene pool. Based on the results, measures of in situ conservation and monitoring
of the Taxus baccata population and all identified rare and endangered species at the
locality „Jelak” have been defined.

abstract = "The main long-term goal of conservation and sustainable development of National
Park (NP) „Kopaonik” is to conserve and improve the natural values, including forest
genetic resources. It is located in central Serbia, covering an area of 11,969.04 ha.
One of the 13 localities with the first protection regime is „Jelak” (60.39 ha, 1000-1490
m a.s.l.). This research aimed to identify rare, endangered, relict, endemic, or species
„at-risk” at the locality „Jelak”, as a basis for proposing a program for its conservation.
Its main value is a presence of a mixed forest of yew and fir. By terrain reconnaissance,
a total of 13 forest tree species were recorded, among which Ulmus glabra (rare),
Betula pendula (rare/endangered), Populus tremula („at-risk“), and Taxus baccata
(relict), which indicates a significant diversity. In addition, 14 species were recorded in
the shrub layer, among which Acer heldreichii (endemic) and Acer platanoides (rare/
endangered). English yew (Taxus baccata L.) is a very rare species that occurs only in
the locality „Jelak” in the entire area of NP „Kopaonik”. This species is under varying
degrees of nature conservation in different European countries, and it is categorized
on the European Red List Category and Criteria („least concerning”). Since it is considered a tertiary relict in the forest fund of Serbia, it is important to conserve the available gene pool. Based on the results, measures of in situ conservation and monitoring
of the Taxus baccata population and all identified rare and endangered species at the
locality „Jelak” have been defined.",

AB  - Cilj rada bio je utvrđivanje varijabilnosti dela genofonda hrasta kitnjaka sa područja Spomenika prirode "Šuma Košutnjak" na nivou jedno- i dvogodišnjih sadnica, u testu potomstva osnovanom u rasadniku Šumarskog fakulteta. Istraživanje je sprovedeno na jednogodišnjim i dvogodišnjim sadnicama deset linija polusrodnika hrasta kitnjaka. Mereni su visina i prečnik u korenovom vratu, utvrđeno je preživljavanje sadnica na kraju drugog perioda rasta, kao i godišnji visinski i debljinski prirast. Prikazani su rezultati deskriptivne statistike, jednofaktorijalne analize varijanse i Fišerovog testa najmanje značajnih razlika, kao i multivarijantnih analiza - klaster i analize glavnih komponenti. Dobijeni rezultati ukazuju na zadovoljavajući nivo genetičke varijabilnosti, koji predstavlja dobru polaznu osnovu za realizaciju ex situ konzervacije dela genofonda, osnivanjem poljskog ogleda na području Košutnjaka i/ili unošenjem sadnica u manjim ili većim grupama na ovo područje. Na ovaj način, deo srodnika, trajno se čuva, povećava se brojnost polazne populacije i stvaraju se uslovi za kontinuiran naučno-istraživački rad.
