AB  - Phenols are considered priority pollutants of wastewaters, they are non-biodegradable and the problem of their
removal is a current and important enviromental issue. Enzymatic treatment of wastewaters using peroxidase
and H2O2 is highly effective and selective method. Soybean hulls offer cheap source of large amounts of crude
enzyme (soybean hull peroxidase - SHP), which can be used for this purpose. The aim of this research was to
test different macroporous carriers for immobilization of soybean hull peroxidase. Our data demonstrate that
SHP immobilization using glutaraldehyde activation is influenced by the pore size of the macroporous glycidyl
methacrylate matrix. Both specific activity of the immobilized enzyme and immobilization yeald were increased
with the increase in pore size, and the highest specific activity obtained was 22,8 U/g of carrier.

AB  - Removal of phenolic compounds from wastewaters has been investigated by many researchers and it has been shown that horse radish peroxidase can react with aqueous phenolic compound in the presence of hydrogen peroxide and form non-soluble materials that could be easily removed from the aqueous phase. Main disadvantage of this process is enzyme inactivation. In order to increase stability of enzyme and yield of biocatalytic reaction peroxidases were immobilized on different materials. The aim of this study was to test different macroporous glycidyl methacrylates as carriers for enzyme immobilization. Immobilization was performed using carriers with different pore sizes (40, 50, 120 and 200 nm) and glutaraldehyde as activation method. All immobilized enzymes were characterized using pyrogallol and H2O2 as substrates (13 mM and 10 mM, respectively), and specific activity and immobilization yield were determined. Our data demonstrate that the best carrier for immobilization of horse radish peroxidase was SGE EDA 20/12 with pore size of 120 nm.

Fenoli se smatraju najvećim zagađivačima otpadnih voda, konstantno prisutni u životnoj sredini i njihovo uklanjanje se može izvesti i enzimskim metodama. Iako je primena enzima, kao što su peroksidaze, bio predmet velikog broja istraživanja, glavni nedostatak i dalje predstavlja visoka cena prečišćenog enzima. Imobilizovani enzimi, sa druge strane, imaju neke prednosti, kao što su povećana stabilnost i lako odvajanje iz reakcionog medijuma, što omogućava višestruku upotrebu. Cilj ovog rada bio je optimizacija uslova za
imobilizaciju peroksidaze iz soje na makroporozni glicidil metakrilat aktiviran glutaraldehidom.
Imobilizacija je rađena u malim tubama, mešanjem 1 ml rastvora enzima (0,034-1,25 mg/ml) sa 50 mg matriksa (makroporozni glicidil-metakrilat prethodno sintetisan u našoj laboratoriji) na rotacionoj mešalici (24h na 25°C, 250 rpm). Enzimska aktivnost merena je koristeći pirogalol i vodonik peroksid kao supstrate (13 mM i 10 mM, redom), a reakcioni proizvodi praćeni su merenjem apsorbancije na 420 nm, tokom 3 minuta.
Naši podaci pokazuju da peroksidaza iz soje može uspešno da se imobilizuje na glutaraldehidno aktiviran makroporozni glicidil-metakrilatni nosač i najbolji imobilizat je imao specifičnu aktivnost od 229 U/g nosača.

AB  - Phenols are considered priority pollutants of wastewaters, persistently present in the environment and its conventional treatment could be overcome using enzymatic methods. Although the application of enzymes, such as peroxidases, has been investigated widely, its main disadvantage is the cost of purified enzymes. Immobilized enzymes, on the other hand, have the advantage of greater stability and easy separation from the reaction medium, which makes them reusable. The aim of this study was to optimize the conditions for soyabean peroxidase glutaraldehyde immobilization technique.
Immobilization was done in small tubes, by mixing 1 ml of enzyme solution (0.05-2.5 mg/ml) with 50 mg of support matrix (macroporous glycidyl methacrylates previosly synthesized in our lab) on a rotary shaker
(24h at 25°C, 250 rpm). Enzyme activity was measured using pyrogallol and H2O2 as substrates (13 mM and 10 mM, respectively), and the reaction products were measured following absorbance at 420 nm, for 3 min
using a spectrophotometer. 
Our data demonstrate that soyabean peroxidase can be successfully immobilized on macroporous glycidyl methacrylate using glutaraldehyde activation.
