The Effect of Biotic Stress in Plant Species Induced by ‘Candidatus Phytoplasma solani’—An Artificial Neural Network Approach

AB  - Infections with phytoplasma present one of the most significant biotic stresses influencing
plant health, growth, and production. The phytoplasma ‘Candidatus Phytoplasma solani’ infects
a variety of plant species. This pathogen impacts the physiological and morphological characteristics of plants causing stunting, yellowing, leaf curling, and other symptoms that can lead to significant economic losses. The aim of this study was to determine biochemical changes in peony (Paeonia tenuifolia L.), mint (Mentha × piperita L.), and dill (Anethum graveolens L.) induced by ‘Ca. Phytoplasma solani’ in Serbia as well as to predict the impact of the biotic stress using artificial neural network (ANN) modeling. The phylogenetic position of the Serbian ‘Ca. Phytoplasma solani’ strains originated from the tested hosts using 16S rRNA (peony and carrot strains) and plsC (mint and dill strains) sequences indicated by their genetic homogeneity despite the host of origin. Biochemical parameters significantly differed in asymptomatic and symptomatic plants, except for total anthocyanidins contents in dill and the capacity of peony and mint extracts to neutralize superoxide anions and hydroxyl radicals, respectively. Principal Component Analysis (PCA) showed a correlation between different chemical parameters and revealed a clear separation among the samples. Based on the ANN performance, the optimal number of hidden neurons for the calculation of TS, RG, PAL, LP, NBT, •OH, TP, TT, Tflav, Tpro, Tant, DPPH, and Car was nine (using MLP 8-9-13), as it produced high r2 values (1.000 during the training period) and low SOS values. Developing an effective early warning system for the detection of plant diseases in different plant species is critical for improving crop yield and quality.

A contribution to the estimation of berry fruits quality

AB  - This study aimed to estimate the quality (in terms of standard chemical parameters, total antioxidative activity /AO/ and thermal properties) of the fresh strawberry (Fragaria xananassa L. Duch.), raspberry (Rubus idaeus L.) and blackberry (Rubus fruticosusL.) samples grown in the Western Serbia. The examined blackberry variety was found to exhibit the most potent AO (DPPH /2,2-diphenyl-1-picrylhydrazyl/ assay, EC50 4.05 +/- 0.22 mu g and ARP 0.25 +/- 0.03 mu g(-1); Hydroxo-Perhydroxyl Mercury(II) Complex /HPMC/ assay, 0.072 +/- 0.007%/mu L), due to the enriched contents of both total phenolics and anthocyanins. It was actually followed by the raspberry and strawberry samples. Unlike the aforementioned trend for AO, thermal behaviour of all samples was similar. This is first time ever that a DC polarographic HPMC assay has been applied for the characterisation of the fresh berry fruits. Further studies aiming to screen total AO of the soft fruit during growth, storage and processing might be well facilitated by the application of this low cost, rapid, easy-to-handle and reliable electrochemical assay.

Antioxidant Activity of Selected Polyphenolics in Yeast Cells: The Case Study of Montenegrin Merlot Wine

AB  - Screens of antioxidant activity (AA) of various natural products have been a focus of the research community worldwide. This work aimed to differentiate selected samples of Merlot wines originated from Montenegro, with regard to phenolic profile and antioxidant capacity studied by survival rate, total sulfhydryl groups and activities of glutathione peroxidase (GPx), glutathione reductase and catalase in H2O2-stressed Saccharomyces cerevisiae cells. In this study, DPPH assay was also performed. Higher total phenolic content leads to an enhanced AA under both conditions. The same trend was observed for catechin and gallic acid, the most abundant phenolics in the examined wine samples. Finally, the findings of an Artificial Neural Network (ANN) model were in a good agreement (r(2) = 0.978) with the experimental data. All tested samples exhibited a protective effect in H2O2-stressed yeast cells. Pre-treatment with examined wines increased survival in H2O2-stressed cells and shifted antioxidative defense towards GPx-mediated defense. Finally, sensitivity analysis of obtained ANN model highlights the complexity of the impact that variations in the concentrations of specific phenolic components have on the antioxidant defense system.
