AB  - BACKGROUND
Fungi absorb and solubilize a broad spectrum of heavy metals such as vanadium (V), which makes them a main route of its entry into the biosphere. V as vanadate (V5+) is a potential medical agent due to its many metabolic actions such as interaction with phosphates in the cell, and especially its insulin-mimetic activity. Antidiabetic activity of V-enriched fungi has been studied in recent years, but the biological and chemical bases of vanadium action and status in fungi in general are poorly understood, with almost no information on edible fungi. 
METHODS
This manuscript gives a deeper insight into the interaction of V5+ with Coprinellus truncorum, an edible autochthonous species widely distributed in Europe and North America. Vanadium uptake and accumulation as V5+ was studied by 51V NMR, while the reducing abilities of the mycelium were determined by EPR. 31P NMR was used to determine its effects on the metabolism of phosphate compounds, with particular focus on phosphate sugars identified using HPLC.
RESULTS
Vanadate enters the mycelium in monomeric form and shows no immediate detrimental effects on intracellular pH or polyphosphate (PPc) levels, even when applied at physiologically high concentrations (20 mM Na3VO4). Once absorbed, it is partially reduced to less toxic vanadyl (V4+) with notable unreduced portion, which leads to a large increase in phosphorylated sugar levels, especially glucose-1-phosphate (G1P) and fructose-6-phosphate (F6P).
CONCLUSIONS
Preservation of pH and especially PPc reflects maintenance of the energy status of the mycelium, i.e., its tolerance to high V5+ concentrations. Rise in G1P and F6P levels implies that the main targets of V5+ are most likely phosphoglucomutase and phosphoglucokinase(s), enzymes involved in early stages of G6P transformation in glycolysis and glycogen metabolism. This study recommends C. truncorum for further investigation as a potential antidiabetic agent.

AB  - Fungi could absorb heavy metals, metalloids, or radionuclides, thus fungal species possess great
potential in bioremediation. Since fungi absorb the vanadium, in the present study ability of Coprinellus
truncorum mycelia for vanadate uptake and its intracellular metabolism were investigated. The
submerged cultivated C. truncorum was exposed to a rising concentration of vanadate. 31P NMR
spectroscopy was used to investigate phosphate metabolism of the mycelium, while the status of
vanadium in the cell was followed by 51V NMR spectroscopy. The mycelium could grow, and overcome
vanadate presence, up to the concentration of 1.6 mM in the submerged medium. 31P NMR
measurements pointed out that vanadate induced changes in the concentration of the crucial metabolite
containing phosphorus, particularly sugar phosphates. The major result of vanadate action is evinced
through an appearance of a signal positioned at around 2.8 ppm, and an increased signal of hexosephosphates.
Using 51V NMR spectroscopy the presence of vanadate monomer in the mycelia of the
fungal cell was confirmed.

AB  - This study was focused on in vitro screening of the total antioxidant activity of the selected extracts of the mushroom Coprinus comatus and quinic acid, one of their antidiabetic ingredients, by an uncommon electrochemical assay. Indeed, direct current (DC) polarographic HydroxoPerhydroxo Mercury(II) Complex (HPMC) assay based on decrease of anodic limiting current originating from HPMC formation in alkaline solutions of hydrogen peroxide at potential of mercury dissolution, observed upon gradual addition of antioxidants, was applied herein for the estimation of the natural products' antioxidativity. Quinic acid was found to exhibit most promising antioxidant potential (4.0 +/- 0.2%mu L-1) being approximate to 2-fold more active than the screened C. comatus extract samples. Actually, such a finding puts some light on the antioxidativity of cyclic polyols, well understimated class of organic compounds, compared to aromatic (poly)phenolics. As a low cost, easy-to handle and accurate this polarographic assay may be thoroughly recommended for much broader use.

AB  - In vitroacetylholinesterase (AChE) inhibitory activity of an autochthonous sample of the mushroomCoprinus comatus(encompassing fruiting body FB, mycelia M and filtrate F from the submerged cultivation) was the subject of this study.C. comatusF extract exhibited rather potent anti-AChE activity (73.0 +/- 1.5%) atin liquidconditions, comparable to those of the conventional drug donepezil (80.6 +/- 1.4%). Also, the same extract exhibited high anti-AChE activity (1 mu g)in solid. While its FTIR spectrum indicated the presence of phenolic compounds, quercetin (28.1 mu g g(-1)d.w.) was found to affect the observed bioactivity (59.8 +/- 0.9%). This is the first report of profound anti-AChE activity of anyC. comatusextract, a medicinal mushroom that has been successfully cultivated in P.R. China, due to the demanding needs of food industry.

AB  - The polysaccharide (PSH) extracts from the edible mushroom species Coprinus comatus and Coprinellus truncorum were screened in liquid for their acetylcholinesterase inhibitory (AChE) activity. Both extracts were found to display inhibition of the aforementioned enzyme reaching similar IC50 values of 0.62 +/- 0.07 and 0.61 +/- 0.03 mg/mL, respectively. According to the means of FTIR spectroscopy, these PSH extracts mostly contained beta-glucans. However, the presence of some proteins and polyphenolics as minor ingredients were also detected. Compared with existing literature data for anti-AChE activity of the sugar samples, the findings within this study may be treated as a profound bioactivity. Consequently, this study puts some light on the possible use of the screened macrofungi in the palliative treatment of Alzheimer's disease. [GRAPHICS] .

AB  - This study outlines antioxidant and anti-AChE activities of the polysaccharide (PSH) extract from the mushroom species Amanita strobiliformis. Both the presence of alpha and ss glucans within the aforementioned extract was recorded. PSH extract displayed a profound scavenging activity of OH radicals (IC50 value, 11.86 +/- 0.59 mu g/mL) and high potential for reduction of Fe3+ ions (174.11 +/- 8.70 mg eq. AA/g d.w.) being almost 48- and 5-fold more effective than mannitol and butylated hydroxytoluene used as a positive control, respectively. Compared with galanthamine (0.001 mu g), the same extract exhibited a moderate anti-AChE activity (10 mu g) in solid. Since purified PSH extract exhibited higher bioactivity (IC50 value 7.27 +/- 0.31 mu g/mL, 197.68 +/- 9.47 mg eq. AA/g d.w. and 0.1 mu g, respectively), it can be predominantly ascribed to the polysaccharide compounds. A. strobiliformis PSH extract may be considered as a promising resource of potent bioactive polysaccharides of natural origin successfully addressing both oxidative stress and lack of acetylcholine.
